انگشت‌نگاری گونه‌های جنس آژیلوپس (Ageilops) با نشانگرهای مبتنی بر ژن‌های هدفمند و نواحی حفاظت شده CoRAP

چکیده

گونه ­های جنس آژیلوپس (Ageilops) از گرامینه­ های مرتعی و یکی از مهمترین اجداد گندم و منبع غنی از ژن­های تحمل به تنش­ هاست. بدین‌منظور انگشت‌نگاری روابط ژنتیکی توده ­های متعلق به هشت گونه ­ Ageilopsبا استفاده از نشانگرهای مولکولی CoRAP مبتنی بر ژن­های هدفمند مورد مطالعه قرار گرفت. در طراحی آغازگرهای ثابت از شش ژن عامل تحمل تنش ­های غیرزیستی CAT، MnSoD، SoS1، miR398، miR160b و miR169gR استفاده شد. کمترین و بیشترین محتوای اطلاعات چندشکلی به­ترتیب مربوط به آغازگرهای CoRAP2 و CoRAP10 با مقادیر 92/0 و 96/0 بود. شاخص نشانگری در بین نشانگرهای مورد بررسی از 89/6 در آغازگر CoRAP7 تا 49/13 در آغازگر CoRAP9 متغیر بود. بین گونه­ ها جریان ژنی (Nm) پایین و بعکس میزان تمایز (Gst) بالایی مشاهده شد. همچنین مقدار شاخص Fst برابر با 45/0 نشان داد که جمعیت­ های مورد مطالعه کاملاً از هم متمایز شده­ اند. با توجه به بالا بودن شاخص­های تعداد آلل مؤثر و تنوع ژنتیکی Nei، دو گونه Ae. cylandrica و Ae. caudata تنوع درون گونه­ ای بالایی در بین گونه ­های مورد بررسی نشان دادند. بیشترین تشابه بین دو گونهAe. truncialis  و Ae. umbelulata و بین دو گونه Ae. neglecta و Ae. cylanrica مشاهده شد. تجزیه خوشه­ ای به‌طور مناسبی گونه­ ها را در گروه ­های مجزا تفکیک و تجزیه به مختصات اصلی نتایج تجزیه خوشه­ ای را تأیید کرد. با تجزیه ساختار جمعیت، نحوه جریان ژنی و اختلاط ژنتیکی بین گونه­ ها مشخص شد که در تطابق با نتایج تجزیه خوشه ­ای و نمودار دوبعدی PCoA بود.
