In vitro propagation of Taxus baccata through zygotic embryo culture

Abstract

Taxus baccata masses are considered to be the oldest forests with the highest density in the world. These experiments have been accomplished to decrease dormancy and increase the germination of Taxus baccata seeds.
The mature seeds from selected trees in natural plant nursary located in Astara forests (North of Iran), were collected in autumn and were laid under different treatments with concentrated HCl and continuous washing by water. After the seeds were washing under running tap water for 15 days, were disinfected by 3% sodium hypochloride  for 15 minutes and then with 0.1% Hgcl2. for 2 minutes. The embryos were aseptically dissected and transferred in modified MS medium. After 4 weeks 65% producted seedlings. Seedlings were transferred to soil in different stages of acclimatization
