The determination of water soluble proteins characteristics in seven forage species using SDS-PAGE technique

Abstract

In order to determine water extract proteins and their effects on ruminal degradation, alfalfa hay, clover, sainfoin, salad crop and corn silage along with two range forage species including Lolium prenne and Glyceria plicata fries (GPF), were used. Extractions were made with deionized water and soluble proteins in the extract were precipitated using 75% (v/v) acetone solution. The precipitated proteins were separated by SDS-PAGE. The insoluble residues along with whole samples were used for ruminal degradation using gas production technique. The highest soluble protein percentage belonged to the salad crop and the lowest percentage was belonged to GPF forage (P<0.05). Non significant difference between whole samples and insoluble residue samples suggested that the value of soluble proteins as a factor for dry matter degradability is limited.
