Background and Objectives: Black nightshade (Solanum nigrum L.), a medicinal species belonging to the Solanaceae family, has attracted considerable attention because of its production of steroidal glycoalkaloids, particularly solasonine, which exhibits remarkable antitumor, anti-inflammatory, and antimicrobial activities. Solasonine glycosyltransferase (SGT1) catalyzes the final step of the solasonine biosynthetic pathway by transferring a glycosyl moiety to the steroidal aglycone solasodine. Despite the biological and pharmaceutical importance of solasonine, limited information is available regarding the SGT1 coding gene and its expression profile in S. nigrum. Therefore, this study aimed to isolate the full-length coding cDNA of the SGT1 gene and investigate its expression pattern in different plant tissues following methyl jasmonate treatment, a well-known elicitor and key regulator of plant secondary metabolism.
Materials and Methods: To evaluate the effect of methyl jasmonate on SGT1 gene expression, a completely randomized design with three replications was employed. Solanum nigrum seeds were grown under greenhouse conditions, and leaf, immature green fruit, and ripe fruit samples were collected one day after spraying reproductive-stage plants with 100 µM methyl jasmonate. Based on transcriptomic data retrieved from the NCBI database and sequence analysis using the BLASTn algorithm, a consensus sequence was generated, and gene-specific primers were designed to amplify the coding cDNA of SGT1. The amplified PCR product was purified, cloned into the pNovel T/A Lethal Vector, transformed into Escherichia coli DH5α competent cells, and subsequently sequenced. Relative gene expression was quantified using quantitative real-time PCR (qRT-PCR) with gene-specific primers, while the Elongation factor 1α gene served as the internal reference. Relative transcript abundance was calculated using the 2^−ΔΔCt method. Statistical analyses, including analysis of variance and Duncan’s multiple range test, were performed using MSTAT-C and Microsoft Excel software.
Results: A full-length coding cDNA of the SGT1 gene, comprising 1,479 base pairs, was successfully isolated and cloned from S. nigrum. Protein sequence analysis identified the highly conserved uridine diphosphate glycosyltransferase (UDPGT) signature motif together with the characteristic plant secondary product glycosyltransferase (PSPG) domain located at the C-terminal region. The conserved HCGTNS motif, recognized as a diagnostic sequence of SGT1 enzymes in the genus Solanum, was identified, while substitution of the conserved tryptophan residue with threonine represented a distinctive structural characteristic of S. nigrum SGT1. Phylogenetic analysis demonstrated that the SGT1 protein of S. nigrum clustered most closely with that of Solanum dulcamara, indicating a close evolutionary relationship between the two species. Gene expression analysis revealed significant effects of tissue type, methyl jasmonate treatment, and their interaction. Overall, methyl jasmonate significantly enhanced SGT1 transcript accumulation compared with untreated control plants. The highest expression level (8.76) was detected in methyl jasmonate-treated immature green fruits, representing a 3.7-fold increase relative to the corresponding control, whereas the lowest expression level was observed in ripe fruits. These findings indicate that methyl jasmonate effectively induces SGT1 expression in all examined tissues, although the magnitude of induction differs among tissues, demonstrating a clear tissue-specific transcriptional response.
Conclusion: The successful isolation and characterization of the SGT1 gene provides valuable insight into the molecular basis of solasonine biosynthesis in S. nigrum. Given the considerable pharmaceutical potential of solasonine, particularly its antitumor properties, the findings of this study provide a valuable foundation for future metabolic engineering and biotechnological strategies aimed at enhancing solasonine production in cell cultures and transgenic plants. Furthermore, elucidating the regulatory effect of methyl jasmonate on SGT1 expression contributes to a better understanding of secondary metabolite biosynthesis and may facilitate the development of effective approaches for improving the production of valuable bioactive compounds in S. nigrum.

Background and Objectives: The genera Alyssum L. and Malva L., belonging to the Brassicaceae and Malvaceae families, respectively, comprise annual and perennial plant species with considerable medicinal and ecological importance. These genera are widely distributed throughout Iran and have long been used in traditional medicine because of their expectorant, antitussive, and laxative properties. However, cytogenetic information regarding the chromosome number, ploidy level, and karyotype characteristics of native populations remains limited or incomplete. Therefore, the present study aimed to determine the somatic chromosome number, assess ploidy diversity, and characterize the karyotypic features of selected populations of Alyssum and Malva in order to provide fundamental cytogenetic information for taxonomic studies, conservation, and future breeding programs.
Materials and Methods: Seeds of eight populations representing Alyssum stapfii, A. hirsutum, Malva sylvestris, and M. parviflora (two populations of each species) were obtained from the Gene Bank of Natural Resources of Iran and grown under controlled conditions following appropriate seed germination treatments. Root-tip meristems were collected for chromosome preparation using the squash technique after pretreatment, fixation, hydrolysis, and hematoxylin staining. Chromosome observations were performed using an Olympus BX40 light microscope, and high-quality metaphase plates were photographed for subsequent analyses. Chromosomal parameters, including long arm length (LA), short arm length (SA), and total chromosome length (TL), were measured using Micro-Measure software. Cytogenetic indices, including the centromeric index (CI), intrachromosomal and interchromosomal asymmetry indices (A1 and A2), total form percentage (TF%), arm ratio (AR), difference in relative chromosome length (DRL), symmetry index (SI), centromeric gradient (CG), and dispersion index (DI), were calculated using Microsoft Excel and standard cytogenetic formulas. Karyotype symmetry was classified according to the systems proposed by Stebbins and Levan. Statistical analyses were performed using SAS software.
Results: In the genus Alyssum, the basic chromosome numbers were x= 7 and x= 8, with tetraploid (2n=4x = 32) and hexaploid (2n=6x = 42 and 48) cytotypes identified among the studied populations. According to Stebbins’ karyotype classification, all Alyssum populations were assigned to symmetry class 1C, with metacentric chromosomes predominating. Analysis of variance revealed that only the centromeric gradient (CG) differed significantly among populations (P≤ 0.01), whereas the remaining cytogenetic parameters showed no significant differences. Based on the values of A1, A2, TF%, SI, and the karyotypic formula, population 30546 (Khalkhal) of A. stapfii exhibited the most symmetrical karyotype. Mean comparison indicated that the greatest total chromosome length (1.31 μm) was recorded in population 23372 (Taft) of A. stapfii. In the genus Malva, the basic chromosome numbers were x=7 and x=14, with hexaploid chromosome complements of 2n=42 and 2n=84 observed among the populations. The Malva populations were classified into Stebbins’ symmetry classes 1B, 1C, and 2C, with metacentric chromosomes predominating in all populations. Significant differences were detected among populations for DRL, A2, and DI (P ≤ 0.01). Among the evaluated populations, population 012 (Tangestan) of M. parviflora exhibited the greatest total chromosome length (1.22 μm).
Conclusion: The results of this study demonstrate substantial cytogenetic diversity among the evaluated populations of Alyssum and Malva, particularly with respect to chromosome number, ploidy level, and karyotype structure. Such diversity is likely associated with adaptation to the diverse ecological conditions of their natural habitats. The cytogenetic information generated in this study provides a valuable foundation for taxonomic revision, evolutionary and phylogenetic investigations, germplasm conservation, and the development of breeding programs for these medicinally important genera.

Background and Objectives: Mahaleb cherry (Cerasus mahaleb L.) belonging to the Rosaceae family is a valuable native tree species of the Irano-Turanian region. Owing to its high drought tolerance and remarkable ability to establish under harsh environmental conditions, it plays an important role in the restoration and conservation of semi-arid mountainous forests. In addition, it contributes to soil stabilization, enhances biodiversity, and serves as a resilient rootstock for fruit trees in horticultural systems. Despite its ecological and economic importance, limited information is available on the phenotypic performance of different genotypes grown under uniform environmental conditions within a common seed orchard. Therefore, this study aimed to investigate the influence of maternal habitat characteristics on the growth and productivity traits of elite mahaleb genotypes established in the Fereydunshahr seed orchard.
Materials and Methods: A total of 120 cuttings representing elite genotypes selected from six natural habitats with contrasting ecological conditions, including differences in elevation, slope, aspect, and soil properties, were evaluated over eight consecutive years in the Fereydunshahr seed orchard. Two important vegetative traits, namely plant height (cm) and canopy diameter (cm), were measured twice annually (spring and summer) during the final three years of the evaluation period. The average values recorded traits in the third year were used as the principal criterion for identifying superior genotypes. In addition, the relationships between vegetative growth traits and ecological characteristics of the maternal habitats, including annual precipitation, temperature, slope, and altitude, were examined using Pearson’s correlation coefficient. Furthermore, hierarchical clustering analysis using Ward’s method was performed to classify the 10 superior genotypes based on their phenotypic performance.
Results: The evaluation of mahaleb genotypes under uniform environmental conditions in the Fereydunshahr seed orchard revealed considerable variation in plant height and canopy diameter over the three-year assessment period. Significant phenotypic differences were detected among the evaluated genotypes, indicating that both genetic and environmental factors of maternal habitats substantially influenced vegetative growth. A very strong positive correlation was observed between plant height and canopy diameter (r≈ 0.96), reflecting a coordinated pattern of vertical and horizontal growth among the superior genotypes. However, there were no significant relationships between either trait and ecological characteristics (annual precipitation, temperature, slope, and altitude. Hierarchical cluster analysis using Ward’s method separated the selected genotypes into two clearly distinguishable groups. Genotypes assigned to Cluster 1 exhibited significantly greater plant height and canopy diameter than those belonging to Cluster 2. These results indicate that, despite cultivation under identical orchard conditions, the genetic background of the genotypes and the environmental characteristics of their maternal habitats continued to exert a pronounced influence on growth performance. The findings provide valuable information for the establishment of improved seed orchards and for the identification of suitable parental genotypes in future breeding programs.
Conclusion: For the establishment of improved seed orchards, it was recommended to combine genotypes exhibiting vigorous vegetative growth with those demonstrating stable and consistent reproductive performance. Such a breeding strategy can facilitate the production of high-quality seeds that are well adapted to diverse environmental conditions. Forest stands established from these improved seeds are expected to possess greater genetic diversity, enhanced adaptability, and increased resilience to environmental stresses, thereby contributing to the long-term sustainability of forest restoration and conservation programs.

Background and Objective: Cocksfoot (Dactylis glomerata L.) is a major cool-season perennial forage grass widely distributed in the rangelands of Iran and extensively used for livestock grazing and forage production. In grass breeding programs, knowledge of genetic variation is essential for achieving sustainable biomass production, broad adaptation, and long-term genetic improvement. Genetic diversity is the cornerstone of grass breeding because it enables effective parental selection, facilitates the exploitation of heterosis in polycross and synthetic varieties, and enhances yield stability and tolerance to biotic and abiotic stresses. Therefore, this study was conducted to investigate the genetic diversity of superior cocksfoot (Dactylis glomerata L.) genotypes using agronomic traits and ISSR molecular markers.
Materials and Methods: Twenty superior genotypes, previously selected based on forage yield, flowering date, and resistance to rust disease, were evaluated for both agronomic traits and ISSR molecular markers. The field experiment was conducted at the research farm of the Agricultural Biotechnology Research Institute, Tabriz, Iran, in 2020. Fifteen clonal plants from each genotype were established in compost-filled pots. After growth under greenhouse conditions, the seedlings were transplanted to the field. The experiment was arranged in a randomized complete block design with three replications. In each plot, five plants of each genotype were established at a spacing of 60 × 60 cm. No data were collected during the establishment year. Data on heading date, pollination date, plant height, panicle length, tiller number, crown diameter, seed yield, and forage dry matter yield at the first and second harvests were recorded in 2022. The data were subjected to analysis of variance, and mean comparisons were performed using the LSD test at the 5% probability level. Agronomic data were further classified using Ward’s cluster analysis. For molecular analysis, 29 ISSR primers were employed. Band presence or absence was scored, and genetic diversity parameters, including the number of effective alleles (Ne), Shannon’s information index (I), and expected heterozygosity (He), were estimated. Genetic relationships among genotypes were evaluated using the Neighbor-Joining clustering method and principal component analysis (PCA).
Results: The results of the field experiment revealed significant differences among genotypes for all measured traits. Genotypes Dg1, Dg6, Dg12, Dg13, and Dg18 exhibited superior forage dry matter and seed yields compared with the remaining genotypes. Cluster analysis based on agronomic traits classified the 20 cocksfoot genotypes into four distinct groups, containing 3, 8, 4, and 5 genotypes, respectively. Molecular analysis identified 346 alleles, of which 337 (97.4%) were polymorphic. The number of alleles per primer ranged from 7 to 20, with an average of 11.93 alleles. The mean values of Shannon’s information index, expected heterozygosity, and the number of effective alleles were 0.447, 0.315, and 1.533, respectively, indicating substantial genetic diversity among the evaluated genotypes. Cluster analysis based on ISSR markers also grouped the genotypes into four clusters, with cluster IV exhibiting the highest level of genetic divergence and diversity. The PCA results confirmed the clustering pattern obtained from the molecular data. The genetic similarity matrix, with coefficients ranging from 0.41 to 1.00, suggested that genetically distant clones could serve as suitable parental materials for polycross breeding to maximize heterosis. A comparison of the clustering patterns derived from ISSR markers and agronomic traits indicated that the grouping of genotypes was not fully consistent between the two analytical approaches.
Conclusion: Based on band clarity, reproducibility, and discriminatory ability, ISSR03, ISSR25, ISSR06, ISSR32, ISSR17, and ISSR15 were identified as the most informative markers for assessing genetic diversity in cocksfoot. Furthermore, the lowest Jaccard similarity coefficient (higher genetic distances) was observed between genotype Dg13 and genotypes Dg17, Dg18, and Dg19. Accordingly, genotypes Dg1, Dg6, Dg12, Dg13, and Dg18 demonstrated superior agronomic performance while also belonging to distinct groups with relatively greater genetic distances. These genotypes are therefore recommended as promising parental candidates for polycross or synthetic breeding programs aimed at maximizing heterosis and broadening the genetic base of cocksfoot cultivars.

Background and Objectives: Thymus daenensis Celak. is one of the most valuable medicinal and aromatic plants native to Iran. Excessive harvesting from natural habitats, prolonged drought, and recurrent rangeland fires have seriously threatened many native and endemic thyme populations. Owing to its high adaptability to harsh environmental conditions and its high essential oil content, T. daenensis is considered a promising species for rangeland restoration and cultivation in marginal lands affected by water scarcity and poor soil fertility. Successful domestication, cultivation, and ecological restoration using this species require the identification of appropriate seed storage conditions that maintain seed viability and maximize germination. Since seed longevity is strongly influenced by initial seed quality and storage conditions, determining the optimum storage temperature is essential for both farmers and gene banks to minimize seed deterioration and preserve seed quality during storage.
Materials and Methods: The plant material consisted of seeds collected from two natural populations of Thymus daenensis originating from Fereydunshahr (Isfahan Province) and Lanjrud (Markazi Province), Iran. Seeds were harvested from cultivated plants in September 2022, dried under shade, and stored in paper bags. The effects of storage temperature and storage duration on seed germination characteristics were evaluated using a factorial experiment arranged in a completely randomized design with three replications. Treatments consisted of seven storage periods (0, 1, 2, 3, 4, 15, and 18 months) and three storage temperatures (ambient laboratory temperature, +4°C, and −18°C). Germination percentage, germination rate, mean germination time, seed vigor index, root length, shoot length, and root-to-shoot length ratio were recorded. Data were subjected to analysis of variance, and treatment means were compared using Duncan’s multiple range test in SAS software.
Results: A significant interaction was observed between storage temperature and storage duration for most germination traits. In the Fereydunshahr population, initial germination immediately after harvest was 59%, which increased after three months of storage to 84%, 91%, and 79% under ambient temperature, +4 °C, and −18 °C, respectively. After 18 months of storage, germination percentages were 61%, 81%, and 83% under the respective storage conditions. For short-term storage intended for planting in the following growing season, seeds stored at +4°C exhibited significantly higher germination percentage, seedling length, and seed vigor index than those stored under ambient conditions or at −18 °C. In contrast, for long-term storage (up to 18 months), −18 °C proved to be the most effective storage temperature for maintaining seed quality, whereas ambient temperature resulted in the greatest decline in seed performance. In the Lanjrud population, initial germination was 100%, and no substantial differences were observed among the three storage temperatures during either short-term or long-term storage, indicating excellent seed longevity and storage stability in this population.
Conclusion: The results demonstrated considerable variation in post-maturation behavior and storage response between the two populations of T. daenensis. Dry storage significantly improved seed germination, particularly in the Fereydunshahr population. For short-term storage (approximately three months) intended for sowing in the subsequent growing season, storage at +4 °C is recommended because it provides the highest germination percentage, seedling growth, and seed vigor. Therefore, farmers are advised to store thyme seeds under refrigerated conditions rather than at ambient temperature. For long-term conservation, storage at −18 °C provided slightly better seed viability than storage at +4 °C. Although this difference was relatively small, it may become economically important for large-scale seed production and long-term germplasm conservation. Consequently, −18 °C is recommended for long-term storage in gene banks and commercial seed storage facilities, whereas +4 °C is adequate for routine short-term seed storage and subsequent field planting.

Background and Objectives: Genetic and environmental factors continuously shape the genetic structure of plant populations, generating valuable diversity that can be exploited in plant breeding programs. Accurate assessment of genetic diversity is essential for the efficient selection of parental genotypes and the development of improved varieties. Although morphological traits have traditionally been used to evaluate genetic variation, their application has become increasingly limited because they are strongly influenced by environmental conditions and developmental stages. Consequently, molecular markers are now widely employed alongside morphological traits to provide a more reliable assessment of genetic diversity. Rosa damascena Mill. is one of the most economically important aromatic and medicinal plants, extensively utilized in the food, pharmaceutical, perfumery, and cosmetic industries. Therefore, the development of cultivars adapted to diverse climatic conditions is of considerable importance. The present study was conducted to investigate the genetic diversity of selected R. damascena genotypes using Start Codon Targeted (SCoT) molecular markers and to compare the molecular classification with grouping based on agronomic performance under rain-fed conditions.
Materials and Methods: The genetic diversity of 12 R. damascena genotypes was evaluated using SCoT molecular markers. The plant materials consisted of seven elite genotypes previously selected based on plant height, flower yield, and flower essential oil content under rain-fed conditions, together with five additional genotypes collected from East and West Azerbaijan provinces of Iran. Young leaf samples were collected, and genomic DNA was extracted using a modified CTAB protocol. Polymerase chain reaction (PCR) amplification was performed using 29 SCoT primers, and the amplified DNA fragments were separated by electrophoresis on 5% polyacrylamide gels. Banding patterns were scored as presence (1) or absence (0). Polymorphic information content (PIC), effective multiplex ratio (EMR), and marker index (MI) were calculated using Microsoft Excel. Genetic diversity parameters, including the number of effective alleles (Ne), Shannon’s information index (I), expected heterozygosity (He), and unbiased expected heterozygosity (uHe), together with principal coordinate analysis (PCoA), were estimated using GenAlEx software. Jaccard’s similarity coefficients and cluster analysis were calculated using NTSYS software. In addition, hierarchical clustering based on agronomic traits measured under rain-fed conditions was performed and compared with the clustering obtained from molecular marker data.
Results: Among the 29 SCoT primers evaluated, 13 generated clear and reproducible amplification products. A total of 88 polymorphic alleles were detected across the 12 R. damascena genotypes. Primer SCoT12 produced the highest number of polymorphic alleles (17), whereas primers SCoT46 and SCoT61 each generated only two polymorphic alleles. On average, 6.76 polymorphic alleles were detected per informative primer. Cluster analysis based on Jaccard’s similarity coefficient and the UPGMA algorithm grouped the 12 genotypes into four major clusters at a similarity coefficient of 0.71. Cluster I included genotypes G40, G28, G8, G35, G29, and G6; Cluster II contained genotype G21; Cluster III comprised genotypes GA1 and GA2; and Cluster IV consisted of genotypes GA3, GA4, and GA5. Principal coordinate analysis supported the clustering pattern obtained from the similarity matrix. Based on agronomic traits, including plant height, flower yield, and flower essential oil content, the seven elite rain-fed genotypes were classified into three major groups. Cluster I contained genotypes G6, G8, and G35; Cluster II included genotypes G21, G28, and G29; whereas Cluster III consisted solely of genotype G40.
Conclusion: The results demonstrated substantial genetic divergence between the genotypes maintained at the Research Institute of Forests and Rangelands and those collected from East and West Azerbaijan provinces, suggesting that these materials do not share a recent common genetic origin. Comparison of the clustering patterns derived from SCoT molecular markers and agronomic traits revealed a moderate level of agreement between the two approaches. These findings indicate that combining molecular markers with phenotypic evaluation provides a more comprehensive assessment of genetic diversity and can facilitate the identification of suitable parental genotypes for future Rosa damascena breeding programs under rain-fed conditions.
