شناسایی نشانگرهای آگاهیبخش SSR و EST-SSR مرتبط با مقاومت به زنگ قهوهای در ژنوتیپهای گیاه مرتعی Aegilops tauschii Coss در مرحله گیاهچهای و گیاه کامل
گونهAegilops tauschii ، گیاهی است یکساله و دیپلوئید (2n = 2x = 14, DD) که بهصورت خودرو در دامنه ارتفاعات و یا دشتهای نزدیک به سواحل دریاهای غیرآزاد، از ترکیه تا چین رویش دارد. از آنجا که نواحی شمالی ایران بهعنوان یکی از مهمترین مراکز پیدایش و تنوع این گونه مطرح است، شناسایی منابع مقاومت به زنگ قهوهای (برگ) گندم در این مناطق بسیار اهمیت دارد. در این پژوهش، مقاومت گونه مرتعی آژیلوپس تائوشی (Aegilops tauschii Coss.) نسبت به شش جدایه مختلف عامل بیماری زنگ قهوه ای در مرحله گیاهچه ای و گیاه کامل ارزیابی شد. برای شناسایی نشانگرهای مرتبط با مقاومت گیاهچه به جدایه های مختلف و مقاومت گیاه کامل در مزرعه، از نشانگرهای SSR و EST-SSR که پوشش مناسبی روی ژنوم D گندم داشتند، استفاده شد. تجزیه ساختار ژنتیکی جمعیت، نشان داد که یکصد ژنوتیپ مورد ارزیابی در دو زیرجمعیت متمایز قرار گرفتند. پس از محاسبه ماتریس ضرایب ساختار ژنتیکی و همچنین خویشاوندی در جمعیت (Kinship)، تحلیل ارتباط نشانگر- صفت با استفاده از مدلهای خطی عمومی و مختلط انجام شد. قطعات تکثیری آغازگرهای ریزماهواره Xgwm2، Xgwm44 و Xgwm30 که بهترتیب بر روی کروموزومهای 3D، 7D و 2D و همچنین آغازگرهای SWES186 و SWEW92 از نشانگرهای EST-SSR که بهترتیب بر روی کروموزومهای 2D و 7D قرار داشت دارای ارتباط معنیدار با سطح مقاومت به بیماری زنگ (با ضریب تبیین بالا) بود. با توجه به ارتباط این نشانگرها با مقاومت اغلب جدایه های مطالعه شده، میتوان نتیجه گرفت که بهعنوان نشانگرهای آگاهی بخش، ظرفیت بالایی در گزینش ژنومی و غربالگری سریع ژنوتیپ ها به کمک نشانگر دارند.

شناسایی ریز RNAها و ژنهای هدف مرتبط در گیاه دارویی مرزه خوزستانی
میرناها (microRNAها)، دسته ای از مولکول های تنظیمکننده کوچک و غیر کدکننده هستند که بیان ژن را از طریق تخریب رونویسی یا سرکوب ترجمه تنظیم می کنند. میرناها، در تنظیم گستره وسیعی از فرایندهای متابولیکی و فیزیولوژیکی در گیاهان مشارکت دارند. خانواده نعناع بهویژه مرزه خوزستانی، گیاهان شناخته شده ای از نظر طعم، عطر و خواص دارویی هستند. تاکنون هیچگونه گزارشی از شناسایی میرنا برای گیاه دارویی مرزه خوزستانی (Jamzad khuzistanica Satureja) ثبت نشده است. ازاینرو، در این مطالعه برای پیشبینی میرنا و ژنهای هدفشان در مرزه خوزستانی، از رویکرد محاسباتی مبتنی بر جستجوی همسانی استفاده شد. یونیژن های غیر کدکننده بهعنوان توالی های کاندید پیشساز میرنا در نظر گرفته شدند. در نهایت پس از ارزیابی پارامترهای عمومی درصد باز GC، حداقل انرژی آزاد تاخوردگی (MFE)، شاخص حداقل انرژی آزاد تاخوردگی (MFEI) و ساختار ثانویه 58 میرنا شناسایی شد که از بین آنها با اعمال معیارهای شناسایی اختصاصی گیاهان و پالایش میرناهای پیشبینی شده از چند رونوشت، در نهایت 10 میرنا شناسایی شد. سپس 930 رونوشت هدف با استفاده از وبسایت psRNATarget برای آنها پیشبینی و با استفاده از ابزار BLASTx نرمافزار (v2.6.0) Blast+ NCBI تفسیر کارکردی شد. بررسی ژنهای هدف نشان داد که ژنهای پاسخدهنده اکسین، ژنهای GRAS ((Gibberlic-acid insensitive (GAI), Rspressor of GAI (RGA) and Scarerow (SCR))، ژن AGO2 ( 2 Argonaute) و ژنهای خانواده LAC (Laccase) از اهداف عمده میرناهای شناسایی شده در مرزه خوزستانی هستند. تجزیهوتحلیل غنیسازی مسیر در ژن های هدف نشان داد که مسیر بیوسنتز متابولیتهای ثانویه بهطور معنی داری جزء اهداف میرناهای شناسایی شده هستند. این مطالعه، اولین گزارش از شناسایی میرنا در مرزه خوزستانی بوده که نقش آنها را در تنظیم ژنهای هدف توصیف می کند.

ارزیابی بیان ژن منتون منتول ردوکتاز تحت تاثیر تنش خشکی، شوری و دما در گیاه نعناع فلفلی (Mentha piperita)
نعناع فلفلی (Mentha piperita) یکی از مهمترین گیاهان دارویی تولید کننده متابولیت های ثانویه است. منتول یک جزء مهم از منوترپن های اسانس نعناع فلفلی است که کاربرد گسترده ای در صنایع داروسازی و مصارف صنعتی دارد. این منوترپن ارزشمند توسط آنزیم منتون منتول ردوکتاز(MMR) تولید می شود. رشد گیاه و همچنین بیان این ژن تحت تأثیر عوامل تنش زای محیطی دستخوش تغییر می شود. در این تحقیق ریزوم های گیاه نعناع فلفلی پس از ضدعفونی سطحی، در ظروف حاوی محیط کشت MS با مقادیر 0، 50 و 100 میلیمولار کلرید سدیم و 0، 50، 100 و 150 میلیمولار مانیتول کاشته شدند و بعد در اتاقک های رشد با دماهای 23، 26 و 29 درجه سانتیگراد قرار گرفتند. سه هفته پس از اعمال تنش ها، بیان ژن برگ گیاهان کشت شده با روش Real Time PCR اندازه گیری شد و اطلاعات حاصل آنالیز شد. نتایج بیانگر این بود که بیان ژن منتون منتول ردوکتاز در غلظت های پایین کلرید سدیم کاهش یافته ولی در غلظت 100 میلیمولار نسبت به گیاه شاهد 97 درصد افزایش داشته است. ولی بیان این ژن در غلظتهای بالای مانیتول و همچنین دمای بالا به حداقل سطح خود رسیده است. با توجه به اهمیت نعناع فلفلی در صنایع داروسازی و صنعتی، افزایش تولید منتول با اعمال تیمارهای مختلف حائز اهمیت میباشد. با توجه به نتایج حاصل، ترکیب های تیماری با صفر میلیمولار مانیتول،50 میلیمولار کلرید سدیم و دمای 23 درجه سانتیگراد باعث افزایش 93 درصدی بیان ژن منتون منتول ردوکتاز نسبت به گیاه شاهد شد و پیشبینی میشود که تولید منتول به حداکثر میزان خود برسد.

انگشتنگاری گونههای جنس آژیلوپس (Ageilops) با نشانگرهای مبتنی بر ژنهای هدفمند و نواحی حفاظت شده CoRAP
گونه های جنس آژیلوپس (Ageilops) از گرامینه های مرتعی و یکی از مهمترین اجداد گندم و منبع غنی از ژنهای تحمل به تنش هاست. بدینمنظور انگشتنگاری روابط ژنتیکی توده های متعلق به هشت گونه Ageilopsبا استفاده از نشانگرهای مولکولی CoRAP مبتنی بر ژنهای هدفمند مورد مطالعه قرار گرفت. در طراحی آغازگرهای ثابت از شش ژن عامل تحمل تنش های غیرزیستی CAT، MnSoD، SoS1، miR398، miR160b و miR169gR استفاده شد. کمترین و بیشترین محتوای اطلاعات چندشکلی بهترتیب مربوط به آغازگرهای CoRAP2 و CoRAP10 با مقادیر 92/0 و 96/0 بود. شاخص نشانگری در بین نشانگرهای مورد بررسی از 89/6 در آغازگر CoRAP7 تا 49/13 در آغازگر CoRAP9 متغیر بود. بین گونه ها جریان ژنی (Nm) پایین و بعکس میزان تمایز (Gst) بالایی مشاهده شد. همچنین مقدار شاخص Fst برابر با 45/0 نشان داد که جمعیت های مورد مطالعه کاملاً از هم متمایز شده اند. با توجه به بالا بودن شاخصهای تعداد آلل مؤثر و تنوع ژنتیکی Nei، دو گونه Ae. cylandrica و Ae. caudata تنوع درون گونه ای بالایی در بین گونه های مورد بررسی نشان دادند. بیشترین تشابه بین دو گونهAe. truncialis و Ae. umbelulata و بین دو گونه Ae. neglecta و Ae. cylanrica مشاهده شد. تجزیه خوشه ای بهطور مناسبی گونه ها را در گروه های مجزا تفکیک و تجزیه به مختصات اصلی نتایج تجزیه خوشه ای را تأیید کرد. با تجزیه ساختار جمعیت، نحوه جریان ژنی و اختلاط ژنتیکی بین گونه ها مشخص شد که در تطابق با نتایج تجزیه خوشه ای و نمودار دوبعدی PCoA بود.

تجزیهوتحلیل پروتئوم پیازچه درگونههای Allium stipitatum و A. scabriscapum ایرانی برای شناسایی پروتئینهای با بیان متفاوت
جنس Allium شامل بیش از 920 گونه در سراسر جهان است. ایران زیستگاه اصلی بسیاری از گونه های وحشی Allium میباشد. در این مطالعه از بافت پیازچه دو گونه A. stipitatum و A. scabriscapum از دو زیرجنسMelanocrommyum و Reticulatobulbosa با هدف ارزیابی و شناسایی الگوی بیانی پروتئینهای ذخیرهای حاصل از روش SDS-PAGE استفاده شد. برای شناسایی لکه های پروتئینی و مشاهده تغییرات بیانی دو گونه از الکتروفورز ژل دوبعدی و طیفسنجی جرمیMALDI TOF/TOF MS استفاده گردید. آنالیز ژل های حاصل از الکتروفورز دوبعدی منجر به شناسایی 138 لکه پروتئینی شد. نتایج حاصل بیانگر شباهت اندک میان ژلها و الگوی بیانی متفاوت در بین نمونه ها بود که بر این اساس تعداد کمی لکه مشابه در هر دو ژل حاصل شد. نتایج نشان داد در پروفایل بیانی پروتئین های بافت پیازچه این نمونهها تفاوتهای معنی داری وجود داشت که با استفاده از آزمون T تعداد 9 لکه پروتئینی دارای بیشترین تغییرات معنیدار در سطح احتمال 5% شناسایی شدند و از بین آنها 3 لکه متفاوت با استفاده از روش طیفسنجی جرمی برای شناسایی انتخاب شد. نتایج آزمایش طیفسنجی جرمی و تطبیق داده های حاصل با پایگاه دادههای NCBI و Uniprot و ابزارهای بیوانفورماتیکی مشخص کرد که لکه های مورد آزمایش با پروتئین های Maturase k، Agmatine coumaroyltransferase-1 و یک پروتئین با عملکرد نامشخص بنام Hypothetical Protein مطابقت داشت. یافته های این مطالعه نشان داد با توجه به اینکه نمونه های متعلق به دو گونه A.stipitatum و A. scabriscapum از یک جنس هستند اما الگوی بیانی پروتئینهای ذخیرهای آنها در بافت پیازچه بسیار متفاوت از یکدیگر است.

تنوع کروموزومی و اندازه ژنوم در گیاه داروئی مرزه (Satureja spp.)
گیاه مرزه (Satureja spp.) با داشتن گونههای مختلف و ترکیبات با ارزشی همانند تیمول و کارواکرول، دارای اهمیت خاصی در میان گیاهان دارویی است. در این تحقیق، بذرهای هشت جمعیت از سه گونه مرزه (hortensis S. ، S. mutica و boissieri S.) و همچنین بذرهای دو رقم اصلاح شده آلمانی مرزه (S. hortensis cv. Aromag) و (S. hortensis cv. Saturn) از نظر سیتوژنتیکی ارزیابی شدند. نتایج کاریوتیپی نشان داد که هشت جمعیت از گونه S. hortensis (S1P1-S1P8) دیپلوئید (48x=2n=2) و دو جمعیت از گونه های mutica و S. boissieri (S2P1-S3P1) تتراپلوئید (60x =4n=2) با تعداد کروموزومهای پایه متفاوت بهترتیب 24 و 15 بودند. میانگین طول کروموزومها در تمام جمعیت ها mµ 46/1 (دامنهm µ 71/1-19/1) بود. برای اندازهگیری مقدار ژنوم با استفاده از دستگاه فلوسایتومتری، از فلوروکروم PI و گیاه استاندارد جعفری (Petroselinum crispum, 2C DNA = 4.45 pg) استفاده شد. دادهها در قالب طرح کاملاً تصادفی با پنج تکرار (طول کروموزوم) و سه تکرار (اندازه ژنوم) تجزیه آماری شدند. نتایج تفاوت معنیداری را در سطح احتمال یک درصد بین جمعیتها برای دو آماره نشان داد. میانگین مقدار 2C DNA برای تمام دیپلوئیدها pg 52/3 (دامنهpg 8/3-3/3) و برای دو جمعیت تتراپلوئید pg 38/2 (دامنهpg 51/25-2/2) بود. در این مطالعه مقدار اندازه ژنوم برای اولین بار در گونه های ذکر شده گزارش گردید که در کنار سایر صفات اندازهگیری شده میتواند به غنای دانش کاریولوژیک در جنس مرزه Satureja کمک کند.

بهینهسازی ریزازدیادی دو گونه چویل (Ferulago subvelutina Rech.f. and Ferulago angulata (Schlecht) Boiss) در معرض خطر انقراض و بومی ایران
گیاه دارویی چویل (Ferulago ssp.) متعلق به تیره چتریان و بومی ایران است. گونه های این جنس به دلیل خواب بذر، برداشت بیرویه و تنش های محیطی از قبیل خشکسالی به شدت در معرض انقراض هستند. با در نظر گرفتن این گیاه به عنوان یک گیاه دارویی-مرتعی ارزشمند، تولید و احیای آن امری ضروری محسوب میشود. این تحقیق با هدف بهینه سازی ریزازدیادی دو گونه بومی چویل شامل Ferulago angulata و Ferulago subvelutina انجام شد. بیشترین درصد پینه زایی در گونه F. angulata (100 درصد) با کاربرد ریزنمونه ریشهچه (تحت تیمارهایmg/L NAA 1 + mg/L BAP0، mg/L NAA 5/1 + mg/L BAP0، mg/L NAA 2 + mg/L BAP0) و نیز ریزنمونه برگ حقیقی (تحت تیمارهای mg/L NAA 5/0 + mg/L BAP5/0، mg/L NAA 2 + mg/L BAP1) و در گونه F. subvelutina با کاربرد ریزنمونه برگ حقیقی (تحت تیمارهای mg/L NAA 5/0 + mg/L BAP5/0،mg/L NAA 2 + mg/L BAP1) به ترتیب به میزان 33/72 و 66/71 درصد حاصل شد. بهنحویکه کاربرد توأم هورمونهای سیتوکینین و اکسین در باززایی گونه های چویل مؤثر واقع شد؛ به طوریکه بیشترین باززایی در گونه F. angulata به ترتیب با مقدار 33/53 و 66/46 درصد تحت تیمارهایmg/L NAA 2/0 + mg/L BAP5/0 و mg/L NAA 4/0 + mg/L BAP5/0 بوده و در گونه F. subvelutina با میزان 33/53 و 60 درصد بهترتیب در تیمارهای mg/L NAA 2/0 + mg/L BAP1 و mg/L NAA 4/0 + mg/L BAP1حاصل شد. بهترین تیمار ریشه زایی برای هر دو گونه، محیط کشت MS2/1 شاملmg/L IBA 5/0 انتخاب شد. مرحله سازگاری در مخلوطی از خاک، خاک پیت و ماسه با نسبت وزنی 1:1:1 انجام شد که در مجموع 80 درصد از گیاهان تولیدشده از کشت بافت، در محیط بیرون با موفقیت سازگار شدند.

تنوع سوماکلونال در گیاهکهای باززایی شده از اندامهای ساقه، ریشه و لپه لیلکی ایرانی (Gleditschia capsica Desf.)
حفاظت از ژرمپلاسم گونه های درختی بومی جنگلهای هیرکانی به ویژه لیلکی ایرانی (Gleditschia capsica Desf.)، در اولویت پژوهش قرار دارد. مطالعات مولکولی درختان جنگلی در شرایط درونشیشهای بهویژه در ایران بسیار اندک است. در این پژوهش، تنوع سوماکلونال در گیاهان باززایی شده از ریزنمونه های مختلف لیلکی ایرانی در شرایط درونشیشهای با استفاده از نشانگر مولکولی تکرار توالی ساده (ISSR) بررسی شد. برگ پایه مادری (طبیعی) و ریزنمونه های ساقه، ریشه و لپه بهعنوان ژنوتیپهای مورد مطالعه انتخاب و در محیط کشت MS حاوی 5/0 میلیگرم در لیتر TDZ و 5/0 میلیگرم در لیتر 2,4-D کشت شدند. پس از شش واکشت متوالی، تنوع سوماکلونال گیاهان باززایی شده از این ریزنمونه ها بررسی گردید. مشخصه های درصد چندشکلی (PIC)، تنوع نی و شانون برای بررسی تنوع بین ژنوتیپ ها استفاده شد و بعد با استفاده از الگوریتم UPMGA و تجزیه به مؤلفه های اصلی (PCA) خوشهبندی و تفکیک ژنوتیپ ها انجام شد. با استفاده از 10 آغازگر، درصد چندشکلی بین گیاهان باززاییشده بهمیزان 9/87 درصد بهدست آمد. تجزیهوتحلیل واریانس مولکولی (AMOVA)، 16 درصد تنوع بین ژنوتیپها با پایه مادری را نشان داد. نتایج آنالیز خوشهای بهروش UPGMA، ژنوتیپها را در سه دسته مجزا گروهبندی کرد. بر این اساس ژنوتیپهای پایه مادری و ژنوتیپهای ساقه در یک گروه قرار گرفتند. پایه مادری بیشترین و کمترین ضریب تشابه را بهترتیب با ژنوتیپهای ساقه و ریشه داشت. بنابراین پیشنهاد میشود برای مطالعات کشت بافت لیلکی ایرانی از ریزنمونههای ساقه که تشابه ژنتیکی بالایی با ژنوم مادری دارد استفاده شود. بهطورکلی نتایج نشان داد که تنوع سوماکلونال در بین گیاهان باززاییشده با افزایش تعداد دفعات واکشت افزایش خواهد یافت

بررسی ویژگیهای موفولوژیک و بیوشیمیایی مرتبط با عمرگلجایی در جمعیت های نرگس شهلا (Narcissus tazetta L.) در شرایط آب و هوایی خوزستان
گل نرگس شهلا (Narcissus tazetta) از تیره Amaryllidaceae، ازنظر ویژگی های زینتی و دارویی اهمیت فراوانی در دنیا دارد. هدف از این پژوهش بررسی تنوع 13 جمعیت نرگس از مناطق معتدل و گرمسیری کشور برای یافتن جمعیت های برتر از نظر کیفیت پس از برداشت، با توجه به شاخص های مورفولوژیک و بیوشیمیایی بود. این پژوهش به صورت طرح بلوک های کامل تصادفی با 4 تکرار در مزرعه انجام شد. در این تحقیق، شاخص های مؤثر بر کیفیت گل بریده شامل: تعداد ساقه گل دهنده، طول ساقه، وزن ساقه همراه گل، جذب محلول، کیفیت ظاهری، کلروفیل برگ، پرولین گلچه، کربوهیدراتهای محلول گلچه، فنول کل گلچه، محتوای مالون دی آلدئید گلچه، پروتئین گلچه، فعالیت آنزیمهای پراکسیداز و کاتالاز گلچه اندازه گیری شد. نتایج تجزیه واریانس نشان داد که تفاوت بین جمعیت ها برای کلیه صفات بجز کیفیت ظاهری معنی دار بود. بر اساس تجزیه خوشهای داده های بیوشیمیایی، جمعیت ها در دو گروه اصلی تقسیم شدند. گروه اول شامل جمعیت های کازرون، بهبهان، جهرم، ایلام، شیراز و کرمان بود. در این گروه؛ صفات مهمی هم چون کلروفیل کل، جذب محلول و فنول کل بیشترین مقدار را داشتند. گروه دوم به دو زیر گروه تقسیم گردید. در زیر گروه الف جمعیت های خوسف، قائمشهر و خرم آباد قرار داشتند و در زیر گروه ب اهواز، آبدانان، گچساران و مهران قرار گرفتنتد. زیر گروه الف صفاتی چون محتوای پرولین، مالوندی آلدئید، آنزیمهای آنتی اکسیدانی پراکسیداز و کاتالاز عوامل مؤثر در این گروه بودند. در زیر گروه ب ویژگی های پروتئین کل و کربوهیدراتهای محلول بیشترین مقدار را داشتند. از لحاظ عمرگلجایی جمعیتهای اهواز، آبدانان، گچساران و مهران که بیشترین مقدار پروتئین کل و کربوهیدرات های محلول را در پایان آزمایش داشتند و محتوای پرولین، مالوندی آلدئید، آنزیم های آنتی اکسیدانی پراکسیداز و کاتالاز آنها پس از گذشت 10 روز در سطح پایینی بود به عنوان جمعیتهای برتر معرفی شدند. به طور کلی این ارزیابی نشان داد تنوع مورفولوژیک و بیوشیمیایی بالایی در جمعیتهای بومی نرگس وجود دارد که میتواند در نتیجه تنوع اقلیمی بسیار متفاوت در ایران و هم چنین جهش در این گیاه باشد. در مجموع درک چنین تنوع بالایی در مدیریت و حفاظت ژرمپلاسم این گیاه مفید می باشد و بهنژادگر را در تعیین راهبردهای بهرهبرداری، اصلاح و اهلی سازی و کشت و کار این گیاه یاری میکند.

علف باغ (Dactylis glomerata L.) گیاه علوفه ای و مرتعی چندساله دگرگردهافشان متعلق به تیره گندمیان است که برای احیای مراتع، احداث چراگاه و تولید علوفه مناسب میباشد. این پژوهش با هدف بررسی تنوع ژنتیکی جمعیت های علف باغ با استفاده از نشانگرهای مورفولوژیکی جهت شناسایی ژنوتیپ های پرمحصول اجرا گردید. برای این منظور 25 جمعیت علف باغ براساس طرح آزمایشی بلوکهای کامل تصادفی با سه تکرار در مزرعه تحقیقاتی پژوهشکده بیوتکنولوژی کشاورزی واقع در تبریز، در سالهای زراعی 97-1396 مورد ارزیابی قرار گرفتند. نتایج به دست آمده از تجزیه واریانس، اختلاف معنی داری در صفات مورد بررسی بین جمعیت ها نشان داد که بیانگر تنوع ژنتیکی بالا بین جمعیت ها بود. بر اساس نتایج مقایسه میانگین ها، جمعیت های G2،G5 ،G14 ،G16 ،G18 ،G19 ، G24 و G25 با 312 تا 342 گرم در بوته (معادل 73/8 تا 57/9 تن در هکتار) بیشترین عملکرد علوفه خشک سالیانه و جمعیت های G7، G8، G2 و G16 با 27 تا 35 گرم در بوته (معادل 756 تا 980 کیلوگرم در هکتار) بیشترین تولید بذر به عنوان جمعیت های برتر شناسایی شدند. جمعیت G2 از شاهرود و جمعیت G16 از اصفهان دو جمعیت برتر از نظر عملکرد بذر و علوفه معرفی شدند. تجزیه خوشه ای با استفاده از صفات بررسی شده جمعیتها را در چهار گروه دسته بندی کرد که گروه سه با تعداد هفت جمعیت اکثر جمعیتهای برتر را در خود جا داد. در این مطالعه بیشترین فاصله ژنتیکی مربوط به جمعیتهای گروههای یک و سه بود که برای تولید ارقام ترکیبی بایستی گیاهان والدینی برای تلاقی پلیکراس را از بین این جمعیتها انتخاب کرد.

مقایسه عملکرد علوفه و شاخص سطح برگ در تعدادی ژنوتیپهای داخلی و خارجی یونجه Medicago sativa L
این پژوهش به منظور مقایسه عملکرد علوفه و شاخص سطح برگ ژنوتیپهای مختلف یونجه طی دو سال و برآورد شاخص سطح برگ از طریق وزن برگ و ارتفاع بوته طی سالهای ۱۳۹۰ و 1391 به اجرا درآمد. در این آزمایش 20 ژنوتیپ داخلی (سردسیری و گرمسیری) و خارجی یونجه در سه تکرار در قالب طرح بلوکهای کامل تصادفی در بهار سال 1390 در مزرعه مؤسسه تحقیقات اصلاح و تهیه نهال و بذر کرج کاشته شد. نتایج تجزیه واریانس دو ساله عملکرد علوفه تر و خشک بیانگر تفاوت معنیدار بین میانگین ژنوتیپ ها بود. ژنوتیپهای Bami ×Yazdi، Mesa sersa و KFA17 به ترتیب با 99/41، 84/39 و 15/39 تن در هکتار علوفه تر و 75/9، 41/9 و 16/9 تن در هکتار علوفه خشک بیشترین عملکرد علوفه را دارا بودند. علیرغم تفاوت معنیدار بین ژنوتیپها از نظر نسبت برگ به ساقه، تفاوت معنیداری بین میانگین ژنوتیپهای مورد بررسی از نظر شاخص سطح برگ مشاهده نشد. نتایج نشان داد که امکان برآورد سریع، دقیق و کمهزینه شاخص سطح برگ از طریق صفات وزن برگ و ارتفاع بوته میسر میباشد. در این بررسی، صفت وزن برگ (بویژه وزن تر برگ) نسبت به ارتفاع بوته کارآیی بیشتری در برآورد شاخص سطح برگ از طریق یک مدل خطی (بدون ثابت) با ضریب تبیین بالا (94/0R2 ≥) داشت. چنین نتیجه گرفته شد که در گیاه یونجه هر 100 گرم وزن برگ تر 35/0 متر مربع، هر 100 گرم وزن برگ خشک 51/1 متر مربع و هر 10 سانتیمتر ارتفاع معادل 25/0 متر مربع سطح برگ دارا میباشند.
