The species has a high diversity and widely distributed in Iran, so that from 4 sub species in the world, 3 sub-species are grown in Iran. In this study AFLP markers were used to investigate genetic diversity among 47 samples of 6 motherwort populations and the kinship ties between them. The results showed that 6 selected AFLP primer combinations (MseI /EcoRI) amplified 608 scorable DNA bands, of which 355 were polymorphic (polymorphic 58%). The average genetic similarity of samples was estimated based on the Nei coefficient, 0.26 (0.21-0.30). Analysis of molecular variance (AMOVA) showed that the genetic variation was mainly within populations (57%), but variance among the populations was only 43%. A UPGMA dendrogram based on genetic distance also revealed five major branches in accordance to geographical regions. Maximum and minimum values of genetic similarity of the populations were recorded between “Taleghan 6” and “Taleghan 7” (0.7) and “Dargaz 1” and “Taleghan 7” (0.06) respectively. Data showed a completely different banding pattern of subspecies L. Cardiaca subsp Turkestanicus. of Dargaz region. Coefficient of genetic differentiation among the populations (Gst) and gene flow (Nm), average number of emigration exchanged per generation estimated among the populations, were estimated 0.37 and 0.85, respectively.}, keywords = {Keywords: Genetic relatedness,AFLP,Polymorphism,Gene flow}, title_fa = {بررسی تنوع ژنتیکی برخی از جمعیت‌های گیاه دارویی دم‌شیر (Leonurus cardiac L.) با استفاده از نشانگرهای AFLP}, abstract_fa = {چکیده دم‌‌شیر (cardiaca Leonurus) گیاهیست دارویی از تیره نعنائیان که به دلیل اثرات رقیق‌کنندگی خون و آرام‌کنندگی عضلات قرن­هاست که مورد استفاده قرار می­گیرد. این گیاه در ایران دارای پراکندگی و تنوع بالایی است، به‌طوری‌که از 4 زیر گونه موجود در دنیا، 3 زیر گونه آن در ایران وجود دارد. در این تحقیق به‌منظور بررسی تنوع ژنتیکی 47 ژنوتیپ از 6 جمعیت دم­شیر و تعیین روابط خویشاوندی بین آنها از نشانگر AFLP استفاده شد. چهار جفت ترکیب آغازگری EcoRI/MseI در مجموع 608 نوار قابل امتیازدهی ایجاد نمود که 355 نوار (58 درصد) چند‌شکل بودند. متوسط شباهت ژنتیکی نمونه­های مورد آزمایش بر پایه ضریب Nei، 26/0 (30/0-21/0) تخمین زده شد. تجزیه واریانس مولکولی نشان داد که تنوع ژنتیکی به طور عمده در درون جمعیت­های مورد بررسی (57 درصد) بیشتر از بین جمعیت­ها (43 درصد) بود. دندروگرام بدست‌آمده با استفاده از روش UPGMA، 47 ژنوتیپ دم­شیر را به 5 گروه اصلی تقسیم کرد. نتایج حاصل از ماتریس تشابه به‌ترتیب بیشترین و کمترین تشابه را بین نمونه­های طالقان 6 و طالقان 7 با مقدار 7/0 و نمونه‌های درگز 1 و طالقان 7 با مقدار 06/0 نشان داد. تمامی تجزیه و تحلیل­ها نشان از الگوی نواری کاملا متفاوت زیر گونه turkestanicus Leonurus cardiaca subsp. از منطقه درگز بود. ضریب تمایز ژنتیکی در میان جمعیت‌ها (Gst) به مقدار 37/0 (37 درصد) و برآورد جریان ژنی (Nm)، به طور متوسط تعداد مهاجرت رد و بدل شده در هر نسل در میان جمعیت­ها برابر 85/0 (85 درصد) با فرض تعادل هاردی واینبرگ محاسبه شد. تبادل پایین ژن در بین جمعیت­های دم‏شیر می­تواند به دلیل فاصله زیاد بین رویشگاه­ها باشد. اطلاعات بدست آمده نشان می­دهد که جمعیت درگز می­تواند به‌عنوان یکی از والدها در انجام کارهای اصلاحی و تلاقی با سایر جمعیت­ها و زیرگونه­ها استفاده شود.}, keywords_fa = {واژه‌های کلیدی: ارتباط ژنتیکی,AFLP,چند‌شکلی,جریان ژنی}, url = {https://ijrfpbgr.areeo.ac.ir/article_4999.html}, eprint = {https://ijrfpbgr.areeo.ac.ir/article_4999_7285c835516f95989982994c48799467.pdf} }
