The compounds with cytotoxic effects which have been used for their medicinal properties. Agrobacterium-infected hairy roots are characterized by high growth rate and genetic stability that can beprovided noticeable amounts of the biological material. In this study, production of hairy roots by B. pendula was investigated on different explants, media and A. rhizogenes strains. Growth rate of hairy roots were studied during 20 days and the terpenoids content (betulin & betulinic acid) were assayed by HPLC. Hairy root cultures were established 20 days after infection by infecting bark segments of B. pendula by Agrobactrium strains of C58C1 and LB9402 in WPM medium. Growth of hairy root by lines of C58C1 strain (D and E lines) and one line from LB9402 strain (LB1) were significantly better than others. The most betulin content was observed in the hairy roots, 8 days after culture, which was about 0.47 mg  g-1 dry waight (DW). Furthermore, the most amount of antioxidant activity (75%) was assessed on E line roots, 20 days after cultures. Although the highest content of betulinic acid (0.6 mg g-1 DW ) was obtained by LB1 lines but betulin content and antioxidant activity were considerably less than other hairy root lines. Results indicated that due to higher relative performance, E line of hairy roots could be used for analysis of terpenoids and pharmaceutically important metabolites in future studies.}, keywords = {Antioxidant,Betula pendula,betulin,Betulinic acid,hairy root}, title_fa = {تولید ماده مؤثره دارویی با القاء ریشه مویین در توس (Betula pendula)}, abstract_fa = {گیاه توس (Betula pendula Roth) سرشار از ترپنوئیدهای فعال زیستی ازجمله بتولین و اسید بتولینیک می‎باشد. از این ترکیبات به‌دلیل سمیت سلولی بالا استفاده­های وسیعی در تهیه داروهای مهم می­شود. ریشه‌های موئین تولید شده به‌وسیله آگروباکتریوم با رشد سریع و ثبات ژنتیکی بالا، می‎توانند محل تولید بیشتر این متابولیت‎ها باشند. در این پژوهش، القاء ریشه موئین در گیاه توس با استفاده از ریزنمونه­های مختلف، محیط کشت و سویه‌های مختلف آگروباکتریوم بررسی شد. منحنی رشد ریشه‌های موئین تولید شده در محیط  WPMمایع در مدت 20 روز و میزان اسید بتولینیک و بتولین در آنها به‌وسیله HPLC بررسی شد. ریشه‌های تراریخته تنها با استفاده از سویه اگروباکتریوم تومفاسینس  C58C1و اگروباکتریوم رایزوژنز LB9402، بیست روز پس از تلقیح، در قطعات پوست ساقه و همچنین در محیط کشت WPM تولید شد. دو لاین از سویه C58C1 (E و D) و یک لاین از سویه  LB9402 (LB1) رشد بالاتری نسبت به‌سایر لاین‌ها نشان دادند. بالاترین میزان بتولین در این ریشه‎ها حدود 47/0 میلی‌گرم بر گرم وزن خشک در روز هشتم و بیشترین فعالیت آنتی‌اکسیدانی (75 درصد) در ریشه‌های لاین E در روز بیستم کشت به‎دست آمد. حداکثر میزان اسید بتولینیک 6/0 میلی‎گرم بر گرم وزن خشک در لاین LB1 مشاهده شد. در مجموع، نتایج این تحقیق نشان داد که ریشه‌های موئین لاین E به‌علت عملکرد نسبی بالاتر نسبت به‌سایر لاین‌ها، می‎توانند به‌منظور بررسی ترپنوئیدها و سایر متابولیت‎های دارویی در مطالعات آینده استفاده شوند.}, keywords_fa = {آنتی‌اکسیدان,اسید بتولینیک,بتولین,توس,ریشه موئین}, url = {https://ijrfpbgr.areeo.ac.ir/article_109403.html}, eprint = {https://ijrfpbgr.areeo.ac.ir/article_109403_f43641edd32adf6d8d71c14262039146.pdf} }
