Birch genus (Betula) contains secondary metabolites, especially terpenes which have preventive properties for skin cancer, HIV virus, hepatitis, and malaria. Finding a suitable alternative for pharmaceutical industry is essential due to the limited mother trees and danger of extinction of the mentioned species. Considering the problems associated with crop production and forestry, other methods such as in vitro plantation by applying triggers to obtain the herbal ingredients can be used. The purpose of this research was to evaluate the effects of light and dark on Betulin production in four month calli of in vitro produced B. litwinowii. Explants were prepared by leaves of 25 years old mother plants and 5 years old seedlings during summer from Sangdeh site. They were cultured on WPM plantation medium at light and dark conditions. For analysis of callus production a t- independent test with 5 replicates was used, Analysis of Betulin rate was used using a calibration curve that was obtained by injection of standard solutions using HPLC system. Results showed that light conditions (16 hours light and 8 hours dark) with white light intensity of 1000-1500 lux were essential for callogenesis explants and Betulin quantity was higher in explants prepared from older mother plants.}, keywords = {Betula,light conditions,Secondary metabolites,Tissue culture,WPM medium}, title_fa = {اثر سن پایه و نور برکالزایی توس (Betula litwinowii) و بتولین القاءشده در شرایط درون‌شیشه‌ای}, abstract_fa = {گیاهان به‎علت داشتن متابولیت­های ثانویه، اهمیت جهانی و کاربرد وسیعی در زمینه­های گوناگون دارند. جنستوس (Betula) حاوی متابولیت­های ثانویه به­ویژه ترپن­ها است که خاصیت پیش­گیری‌کننده سرطان پوست و درمان‎کنندگی بیماریHIV، هپاتیت و مالاریا را دارد. یافتن جایگزین مناسب برای داروهای مصنوعی با گیاهان دارویی با توجه به محدود بودن تعداد پایه­ها و در معرض خطر انقراض قرار گرفتن گونه‌های گیاهی مورد نظر امری ضروریست. با مدنظر قراردادن مشکلات مربوط به تکثیر و تولید گیاهان دارویی، زراعی و جنگلی، از روش­های دیگری نظیر کشت­های درون شیشه­ای با اعمال محرک­های تولیدی، برای به‎دست آوردن این مواد مؤثره گیاهی می­توان استفاده کرد. هدف از انجام پژوهش حاضر، ارزیابی تأثیر تیمار نور و تاریکی بر میزان تولید ماده مؤثره بتولین در کالوس­های القا شده چهار ماهه پس از کشت ریزنمونه­های برگ گونه­B. litwinowii در شرایط درون شیشه­ای بود. ریزنمونه­ها از برگ­های جوان پایه­های مادری بالغ (بالای 25 سال) و نهال­های پنج ساله از رویشگاه سنگده فریم در فصل تابستان تهیه و در محیط کشت WPM در شرایط نور و تاریکی کشت داده­ شدند. به‌منظور تجزیه و تحلیل کالوس­زایی از آزمون t مستقل غیر­جفتی با پنج تکرار و برای تجزیه میزان بتولین، از منحنی کالیبراسیون حاصل از تزریق محلول استاندارد با استفاده از دستگاه HPLC استفاده شد. نتایج نشان داد شرایط نوری 16 ساعت روشنایی و 8 ساعت تاریکی با نور سفید و شدت 1000 تا 1500 لوکس برای کالوس­دهی ریزنمونه­ها بسیار ضروری بوده و زمانی که ریزنمونه از پایه مادری مسن­تر تهیه شد، میزان بتولین القاء شده بیشتری (سه­ برابر) حاصل شد. }, keywords_fa = {}, url = {https://ijrfpbgr.areeo.ac.ir/article_101543.html}, eprint = {https://ijrfpbgr.areeo.ac.ir/article_101543_d5277fe7e35894bcd645abd2ea67352d.pdf} }
