تجزیه‌وتحلیل کروموزومی بلوط دارمازو (Quercus infectoria G. Oliver)

چکیده

      دارمازو Quercus infectoria G.Oliver)) یکی از گونه­های با ارزش و اصلی جنگل­های زاگرس است و از نظر زیست­محیطی، اقتصادی، ژنتیکی و دارویی حائز اهمیت است. انجام مطالعات سیتوژنتیکی در گونه­ها و جمعیت­های گیاهی به‎دلیل فراهم کردن اطلاعات کمی روی تاریخچه تکاملی گیاه، تعیین قرابت­های بین گونه­ای و مشخصات کاریولوژیکی، اهمیت فوق­العاده­ای دارد. این مطالعه با هدف تعیین کاریوتیپ و ساختار کروموزومی گونه دارمازو و تنوع کاریوتیپی میان چهار جمعیت آن در غرب ایران (سردشت، مریوان، شینه­قلایی و کاکاشرف) با استفاده از مریستم ریشه و سیستم تجزیه تصویری انجام شد. پس از اجرای مراحل پیش­تیمار، تثبیت، هیدرولیز و رنگ­آمیزی، نمونه­های میکروسکوپی تهیه و خصوصیات کاریوتیپی تعیین شدند. نتایج نشان داد که تعداد کروموزوم­های بیشتر سلول­های مورد مطالعه 24 = x2 = n2 بود اما 25 = x2 = n2 و 26 = x2 = n2 و تریپلوئیدی نیز در برخی از سلول­ها مشاهده شد. تجزیه‌وتحلیل واریانس و مقایسه میانگین بر روی سه تا ده سلول 24 کروموزومی از سه تا هفت پایه مختلف در هر جمعیت انجام شد. تفاوت معنی­دار بین جمعیت­های مورد بررسی از نظر صفات طول کروموزوم، طول بازوی بلند و کوتاه مشاهده شد. شاخص­های تقارن، تقارن نسبی کاریوتیپ را در جمعیت­های مورد بررسی نشان دادند. تجزیه به مؤلفه­های اصلی نشان داد که طول کروموزوم، طول بازوی بلند و کوتاه بیشترین نقش را در ایجاد واریانس داشتند. تجزیه خوشه­اینیز جمعیت­های مورد مطالعه را در دو خوشه مجزا طبقه­بندی کرد. به‎طوری‌که دو جمعیت کاکاشرف و شینه قلایی در یک خوشه و دو جمعیت سردشت و مریوان در خوشه دیگر قرار گرفتند.
