مطالعه الگوی بیان MicroRNAهای دخیل در تحمل تنش خشکی در اجداد و خویشاوندان وحشی و اهلی گندم

miRNA ها، مولکولهای کوچک غیرکدکننده هستند که بیان ژنها را از طریق هضم mRNAیا ممانعت از ترجمه تنظیم میکنند. این مولکولها با تنظیم دقیق بیان ژنها، امکان پاسخ مناسب گیاه را به تغییرات مراحل رشدی یا شرایط محیطی فراهم میکنند. در این تحقیق با استفاده از تکنیک Real Time-PCR الگوی بیان miRNA159، miRNA160 و miRNA398 به همراه ژن هدف miRNA398 (CSD) در شش گونه متعلق به جنسهای تریتیکوم و آژیلوپس در کنار دو رقم گندم نان شاهد حساس (دریا) و متحمل به خشکی (سیروان)، در شرایط عدم تنش (FC=100%) و تنش خشکی (FC=25%) مورد بررسی قرار گرفتند. تجزیه qRT-PCR نشان داد که تحت تنش خشکی هر سه miRNA افزایش بیان معنی داری (P≤0.01) نسبت به شرایط عدم تنش داشتند. در بین گونههای مورد بررسی بیشترین میزان بیان miRNA159 در گونه Ae.tauschii، miRNA160 در گونه T.urartu، miRNA398 در گونه T.durum و ژن CSD در گونه T.durum مشاهده شد. در مقایسه میانگین اثر متقابل گونه و تنش بیشترین درصد افزایش بیان برای هر سه miRNA در شرایط تنش خشکی در سه گونه T.durum، T.urartu و Ae.tauschii مشاهده شد. همچنین بیشترین میزان افزایش بیان CSD در گونههای T.urartu، T.durum و Ae.tauschii بهترتیب با 62/6، 21/6 و 038/6 برابر نسبت به شرایط عدم تنش مشاهده گردید. در جمعبندی مشخص شد که سه گونه T.urartu، T.durum و Ae.tauschii به دلیل برتری از لحاظ miRNAهای عامل ایجاد تحمل میتوانند کاندیدهای مناسبی برای غنی سازی ژرمپلاسم گندم نان و اصلاح برای تحمل تنش خشکی باشند.

ارزیابی تنوع تودههای اسپرس زراعی (Onobrychis vicifolia Scop) از نظر عملکرد و اجزاء عملکرد علوفه در شرایط اقلیمی اصفهان
DOR: 98.1000/1735-0891.1398.27.16.53.1.1575.1606 بهمنظور بررسی تنوع و روابط بین صفات گیاه اسپرس زراعی، آزمایشی روی 20 توده این گیاه با استفاده از طرح بلوکهای کامل تصادفی در سه تکرار، در مزرعه ایستگاه یزدآباد مرکز تحقیقات کشاورزی و منابع طبیعی اصفهان طی سالهای 1393- 1394 انجام شد. نتایج تجزیه واریانس اختلاف معنی داری را میان تودههای مورد مطالعه برای همه صفات به استثناء تعداد گره، تعداد شاخه فرعی، تعداد برگچه و عرض برگ نشان داد. توده پلیکراس با عملکرد علوفه برابر 3179 کیلوگرم در هکتار بهعنوان توده برتر شناخته شد. پس از آن توده 9147 (کرج) با 2965 کیلوگرم در هکتار علوفه، در مقام دوم قرار گرفت. عملکرد علوفه همبستگی مثبت و معنی داری را با تعداد کل ساقه، تعداد شاخه فرعی، تعداد گره، تعداد برگ و وزن خشک ساقه و برگ داشت. سه مؤلفه اول در تجزیه به مولفه های اصلی، 72 درصد از کل واریانس متغیرها را توجیه کردند. تعداد شاخه فرعی، تعداد گره، تعداد برگ، وزن خشک ساقه و برگ و عملکرد تر و خشک علوفه در هکتار عمده ترین نقش را در تبیین مؤلفه اول داشتند. در مؤلفه دوم، ارتفاع ساقه، عرض برگ، درصد برگ و ساقه و نسبت برگ به ساقه و در مؤلفه سوم تعداد برگچه، تعداد کل ساقه، ارتفاع ساقه فرعی و درصد ماده خشک بیشترین اهمیت را در تبیین این مؤلفه ها دارا بودند. تجزیه خوشه ای، 20 توده مورد بررسی را در چهار دسته مختلف قرار داد که اختلافهای چشمگیری از نظر عملکرد علوفه بین گروهها وجود داشت.

بررسی تنوع ژنتیکی در برخی گونههای یولاف (.Avena spp) بومی ایران
DOR: 98.1000/1735-0891.1398.27.28.53.1.1576.1606 در این تحقیق تعداد 49 نمونه ژنتیکی موجود در کلکسیون یولاف بانک ژن گیاهی ملی ایران، متعلق به چهار گونه Avena barbata، A. wiestii، A. fatua و A. sativa مورد بررسی قرار گرفتند. آزمایش به صورت مشاهدهای، در مزرعه پژوهشی مؤسسه تحقیقات اصلاح وتهیه نهال و بذر در کرج انجام شد. صفات آگرومورفولوژیکی مطابق با دستورالعمل مؤسسه ذخایر توارثی گیاهی بینالمللی ارزیابی گردید. براساس مقادیر شاخص شانون، صفت رنگ لما دارای بالاترین مقدار تنوع (42/1) بود. در تجزیه به مؤلفههای اصلی، 43/68 درصد تغییرات موجود در دادهها، توسط هفت مؤلفه اصلی اول توجیه شد.دو نمونه ژنتیکی 439 (A. barbata) و 449 (A. barbata) دارای نزدیکترین و دو نمونه ژنتیکی 447 (A. wiestii) و 115059 (A.wiestii) دارای دورترین فاصله ژنتیکی بودند. نمونههای ژنتیکی مورد مطالعه با استفاده از تجزیه خوشهای به روش K meansدر پنج گروه قرار داده شدند. براساس نتایج تجزیه تابع تشخیص، در مجموع سه تابع تشخیص حاصل گردید که بهمیزان 100 درصد از واریانس کل را شامل میشد. تابع اول بیشترین تمایز را بین دو گونه A. barbata و A.sativa، و تابع دوم بیشترین تمایز را بین گونههای A. fatuaو A. wiestii ایجاد نمود. مجموع نتایج این تحقیق بیانگر وجود تنوع مناسب در جمعیتهای مورد بررسی از لحاظ صفات اندازهگیری شده بود که در امر بهنژادی یولاف میتواند مورد استفاده قرار گیرد. همچنین از توابع تشخیص ایجاد شده در این تحقیق، میتوان برای تمایز بین گونههای مربوطه استفاده کرد.

تأثیر اکسین و برخی ترکیبات سیگنالی بر رشد و تولید متابولیتهای ثانویه در کشت درونشیشهای قرهقاط ( Vaccinium arctostaphylos L)
DOR: 98.1000/1735-0891.1398.27.45.53.1.1578.41 قره قاط یکی از گیاهان دارویی مهم در طب سنتی است که بهمنظور کاهش قند خون و تنظیم فشار خون از دیرباز در ایران کاربرد دارد.در این تحقیق تأثیر اکسین، اسید جیبرلیک، کازئین هیدرولایسیت، پوترسین و سالیسیلیک اسید بر استقرار ورشد ریزنمونه های قره قاط و میزان ترکیبات ثانویه آنتوسیانین، فلاونوئیدی و فنل تام بررسی شد. ریزنمونههای تک گرهی و جوانه های انتهایی قره قاط از رویشگاههای طبیعی تهیه شده و پس از ضدعفونی سطحی روی محیط کشت پایه MS حاوی 2 میلیگرم بر لیتر BAPو سطوح مختلف IBA یا NAA (01/ 0تا 1 میلی گرم بر لیتر) و محیط کشت MS حاوی 2 میلی گرم بر لیتر BAP و 1/0 میلیگرم بر لیتر NAA یا IBA به همراه سطوح مختلف اسید جیبرلیک (0 تا 5/0 میلیگرم بر لیتر)، کازئین هیدرولایسیت (صفر تا 150 میلیگرم بر لیتر)، پوترسین و سالیسیلیک اسید (0 تا 10 میلیگرم بر لیتر) کشت شدند. نتایج نشان داد درصد برگ دهی و زنده مانی ریزنمونه ها بهطور معنیداری تحت تأثیر نوع و غلظت اکسین قرار گرفت. درصد برگ دهی و زنده مانی ریزنمونهها در تیمار هورمونی NAA در غلظتهای پایین تر بهطور معنیداری بیشتر از غلظت های بالاتر NAA و کلیه سطوح IBA بود. اضافه کردن GA3 به ترکیب محیط کشت باعث افزایش معنیدار تعداد برگ در ریزنمونه شد؛ ولی تأثیری در زنده مانی و درصد برگدهی ریزنمونهها نداشت. علاوهبراین کازئین هیدرولایسیت، پوترسین و اسید سالیسیلیک باعث افزایش تعداد برگ و درصد زنده مانی ریزنمونهها گردید ولی این افزایش در برخی از تیمارها از نظر آماری معنیدار نبود.از نظر میزان متابولیتهای ثانویه بین تیمارهای مختلف اختلاف معنی داری وجود داشت. بیشترین میزان ترکیبات آنتوسیانینی در محیط کشت MS+2mg/l BAP+ 0.1mg/l NAA و بیشترین میزان فلاونوئید و فنل کل نیز در همان محیط کشت حاوی 15 میلیگرم بر لیتر سالیسیلیک اسید بدست آمد. بنابراین، از نتایج و دستورالعمل ارائه شده در این تحقیق میتوان در کشت و تکثیر درونشیشهای این گیاه استفاده نمود.

اثر روشهای مختلف پرایمینگ بر بنیه و رشد گیاهچه بذرهای زوال یافته در سه گونه گون Astragalus spp
DOR: 98.1000/1735-0891.1398.27.60.53.1.1575.32 استفاده از تکنیک پرایمینگ، یک روش مؤثر برای افزایش توان جوانه زنی بذر گیاهان مرتعی میباشد. بهمنظور بررسی تأثیر پرایمینگ بذر بر بهبود جوانه زنی بذرهای زوال یافته، در سه گونه گون علوفه ای، آزمایش فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار در سال 1395 در مؤسسه تحقیقات جنگلها و مراتع کشور در شرایط گلخانه اجرا شد. فاکتور A شامل سه گونه از گون (Astragalus cyclophyllon ، A. siliqusus و A. homosus)، فاکتور B شامل دو سطح از زوال بذر، زوال طبیعی (بذرهای نگهداری شده در سردخانه فعال 4 درجه سانتیگراد بهمدت 12 سال) و زوال مصنوعی (با پیری تسریع شده بذرها در دمای 40 درجه سانتیگراد و رطوبت نسبی 100% بهمدت 48 ساعت) و فاکتور C شامل تیمار پرایمینگ بذر در 4 سطح شامل: اسموپرایمینگ (3/0- و 6/0- مگاپاسکال) با استفاده از پلیاتیلن گلایکول PEG6000، هیدروپرایمینگ (خیس کردن بذر در آبمقطر بهمدت 24 ساعت) و شاهد بودند. صفات درصد جوانه زنی، سرعت جوانه زنی، بنیه گیاهچه، طول ریشه و ساقه و گیاهچه، نسبت طول ریشه به ساقه و وزن تر گیاهچه اندازه گیری گردید. نتایج تجزیه واریانس نشان داد که اثرهای اصلی و متقابل فاکتورها برای بیشتر صفات از لحاظ آماری معنیدار بودند. در مقایسه بین گونهها، میانگین صفات در گونه A. siliqususنسبت به دو گونه دیگر بیشتر بود. اثر متقابل پرایمینگ در زوال بذر برای بیشتر صفات معنیدار بود و در زوال طبیعی هیدروپرایمینگ و اسموپرایمینگ 3/0- مگاپاسکال باعث افزایش 25 تا 96 درصدی میانگین کلیه صفات بهجز طول ریشه شد. در حالیکه در زوال مصنوعی هیدروپرایمینگ باعث افزایش 37 درصدی بنیه بذر نسبت به شاهد شد. بهطور کلی نتایج نشان داد که برای احیاء بذر گونهای زوال یافته در بانکهای ژن میتوان از تکنیکهای هیدروپرایمینگ و اسموپرایمینگ 3/0- مگاپاسکال بهعنوان روشی کارآمد در بازیافت بذرهای زوال یافته گون استفاده کرد.

امکانسنجی نگهداری بذرهای شبخسب (Albizia julibrissin Durazz) در شرایط فراسرد
DOR: 98.1000/1735-0891.1398.27.73.53.1.1575.1583 شب خسب یکی از گونه های درختی نیامدار بومی جنگلهای جلگه ای و پایین بند ناحیه هیرکانی است که بهدلیل ابتلا به بیماری قارچی دچار صدمات زیادی شده است. در این تحقیق، امکان استفاده از فناوری فراسرد برای ذخیره سازی بذرهای شب خسب مورد بررسی قرار گرفت. جمعآوری بذر از تعداد 10 پایه درختی از جنگلهای پایینبند حوزه غرب هراز واقع در استان مازندران انجام شد. بهمنظور امکان سنجی قابلیت نگهداری بذرهای شب خسب در شرایط فراسرد از نتایج خصوصیات جوانه زنی بذرهای شب خسب در دو حالت نگهداری در شرایط فراسرد (196- درجه سانتیگراد) با چهار پیش تیمار آماده سازی شامل: ویتریفیکاسیون، گلیسرول 30 درصد، کاهش رطوبت و بدون پیش تیمار به همراه تیمار شاهد (نگهداری در شرایط خشک و دمای 15 درجه سانتیگراد) استفاده شد. بذرها پس از نگهداری یکهفته و یکماه در شرایط فراسرد خارج و در آب 42 درجه سانتیگراد (شوک حرارتی) قرار داده شدند. سپس برای جوانه زنی درون پتریدیش قرار گرفته و به ژرمیناتور با دمای 22+ درجه سانتیگراد منتقل گردیدند. ثبت و شمارش بذرهای جوانهزده بصورت روزانه تا زمانی که دیگر هیچ بذری جوانه نزد انجام شد. نتایج تحلیل واریانس دوطرفه و آزمون مقایسه میانگین دانکن نشان داد که صفات مختلف جوانهزنی بذرهای شبخسب در 4 پیشتیمار مربوط به فراسرد با تیمار شاهد تفاوت فاحشی نداشت. ازاینرو نتیجه گیری شد که نگهداری بذرهای شبخسب در شرایط فراسرد امکانپذیر است. نتایج این تحقیق همچنین تصریح می کند که بذرهای گونه شبخسب به دلیل داشتن پوسته سخت بذر و محتوای رطوبتی بسیار پایین می توانند بدون استفاده از مواد ضدانجماد (بدون پیش تیمار) در دمای 196- درجه سانتیگراد نگهداری شوند. ازاینرو میتوان بدون انجام پیش تیمار نسبت به ذخیره سازی بذرهای شبخسب در شرایط فراسرد اقدام نمود و بانک بذر فراسرد را بهعنوان راهبردی مؤثر برای حفاظت از ظرفیت ژنتیکی این گونه ارزشمند در سطح جنگلهای هیرکانی پیشنهاد کرد

ریزازدیادی اکالیپتوس گونه Eucalyptus citriodora H
DOR: 98.1000/1735-0891.1398.27.88.53.1.32.97 در بین گونههای درختان جنگلی سخت چوب، Eucalyptus citriodoraدرخت سریعالرشدی است که بهدلیل دارا بودن اسانس معطر در عطرسازی، داروسازی و داشتن چوب نرم و سبک در صنایع کاغذسازی و مبلسازی کشت میشود. در این تحقیق، ریزازدیادی E. citriodora به روش کشت جوانه جانبی در محیط کشتهای MS و WPM بررسی شد. سترونسازی با دو محلول کلرید جیوه 1/0 درصد و هیپوکلریت سدیم 20 درصد حجمی اعمال گردید. شاخهزایی با هورمونهای سیتوکینین (BAP، Kin)، اکسین (IBA) و جیبرلین (GA3) در دو سطح محیط کشت در قالب آزمایش فاکتوریل بر پایه طرح کاملاً تصادفی اجرا شد. ریشهزایی نیز با هورمونهای اکسین IBA، NAA و IAA در محیط کشت MS (2/1 نیترات) در قالب طرح کاملا تصادفی انجام شد. آماربرداری از تعداد شاخهها و جوانهها، طول شاخه، سبزینگی، تعداد گیاهچههای ریشهدار شده و تعداد کل ریشه انجام شد. نتایج نشان داد که بهترین تیمار سترونسازی محلول هیپوکلریت سدیم 20 درصد حجمی به مدت 18 دقیقه بود. تکثیر مطلوب و رشد طولی شاخه در محیط کشت MS (2/1 نیترات) و هورمونهای سیتوکینین BAP، Kin، GA3و IBA بهترتیب در غلظتهای (3/0، 2/0، 1/0 و 01/0) و (3/0، صفر، 1/0 و 01/0) میلیگرم در لیتر، بههمراه 200 میلیگرم در لیتر PVP بود. ریشهزایی مطلوب در محیط کشت MS (2/1 نیترات)، با تیمار هورمونی NAA 1 میلیگرم در لیتر و تیمار تلفیقی IAA + IBA 5/0میلیگرم در لیتر بهدست آمد. در نهایت، گیاهچههای ریشهدار شده در شرایط گلخانه سازگار شدند.نتایج حاصل از این تحقیق نشان داد که کشت بافت روش مناسبی برای تکثیر این گونه ارزشمند در زمانی کوتاه میباشد.

اثر نوع ریزنمونه، محیط کشت و تنظیمکنندههای رشدگیاهی بر کالوسزایی و باززایی غیرمستقیم نخودشیرین (Lathyrus odoratus L.)
DOR: 98.1000/1735-0891.1398.27.116.53.1.1575.41 نخود شیرین (Lathyrusodoratus L.) متعلق به خانواده بقولات و یک گیاه زینتی علفی و رونده با گل های معطر است. این گیاه بهعنوان یک منبع ژنتیکی برای صفات مهم مثل مقاومت در برابر تنشها ازجمله تنش گرمایی محسوب میشود. همچنین به منظور حفظ ساختار ژنتیکی آن، تکثیر این گیاه از طریق کشت بافت میتواند مؤثر باشد. به منظور یافتن پروتکل کارآمد شاخه زایی از کالوس؛ اثر نوع ریزنمونه، محیط کشت و تنظیم کننده های رشد گیاهی بر کالوس زایی و شاخه زایی نخود شیرین بررسی شد. نتایج نشان داد که ریزنمونه میانگره نسبت به ریشه کالوس بیشتری تولید کرد (77/0 گرم کالوس برای هر ریزنمونه) و محیط کشت MS نسبت به محیط کشت B5 تأثیر بیشتری در کالوس زایی داشت (78/0 گرم کالوس برای هر ریزنمونه). درمجموع بیشترین کالوس زایی (9/1 گرم کالوس برای هر ریزنمونه) هنگامی که ریزنمونه های میانگره در محیط کشت MS همراه با 5/0 میلیگرم 6- بنزیل آمینو پورین (BAP) و 2 میلیگرم در لیتر 1- نفتالین استیک اسید (NAA) کشت شد، به دست آمد. بیشترین باززایی شاخه (33/5 عدد شاخه برای هر ریزنمونه) از کالوس های کشتشده در محیط کشت MS همراه با 1/0 میلی گرم در لیتر تیدیازورون (TDZ) به دست آمد. این پروتکل می تواند پایه و اساس تحقیقات آینده در ارتباط با بهبود ژنتیکی و تولید انبوه نخود شیرین در کشور باشد.

DOR: 98.1000/1735-0891.1398.27.103.53.1.1575.41 گیاه استویا (Stevia rebaudiana Bertoni) به علت محتوای بالای شیرین کننده های طبیعی قابل استحصال دارای ارزش فوقالعاده بالایی است.با توجه به اهمیت گیاه از نظر صنعتی و بهویژه دارویی، این پژوهش به منظور بهینه سازی جنینزایی بدنی گیاه انجام شد. تولید و القای کالوس رویان زا با استفاده از ترکیبات مختلف تنظیمکننده های رشدی شامل بنزیل آدنین (BA)، نفتالین استیک اسید (NAA) و 2 و 4-دیکلروفنوکسی استیک اسید (2,4-D) در ریزنمونههای برگ و جوانه گیاه در محیط کشت MS به صورت فاکتوریل در قالب طرح کاملا تصادفی در پنج تکرار بررسی شد. بیشترین میزان کالوس زایی در محیط کشت حاوی 01/0 میلی گرم بر لیتر BA + 5/0 میلیگرم بر لیتر 2,4-D و بیشترین میزان رویان زایی در تیمار 01/0 میلیگرم بر لیتر BA +1 میلی گرم بر لیتر 2,4-D حاصل شد. همچنین اثر کازئین هیدرولیزات در پنج سطح (0، 50، 100، 150 و 200 میلی گرم بر لیتر) و شیره نارگیل در دو سطح (صفر و 55 میلیلیتر) بر رویان زایی گیاه استویا توسط دو ریزنمونه برگ و جوانه در آزمایشی به صورت فاکتوریل در قالب طرح کاملا تصادفی با شش تکرار بررسی شد. در غلظت های صفر و 50 میلی گرم بر لیتر کازئین هیدرولیزات به ترتیب بیشترین (2/20 درصد) و کمترین (84/7 درصد) رویان زایی حاصل شد. بیشترین درصد رویان زایی (82/78 درصد) در محیط کشت MS بدون شیره نارگیل توسط ریزنمونه های جوانه و کمترین درصد رویانزایی با 09/25 درصد توسط ریزنمونههای برگ بهدست آمد. پس از آن رویان های حاصل به محیط کشت حاوی 1/0 میلیگرم بر لیتر GA3 (اسید جیبرلیک) برای اندام زایی انتقال یافتند و در این بین 03/18 درصد از رویانها به گیاهچه کامل تبدیل شدند.

بررسی اثر غلطت آنتیاکسیدانهای مختلف برکاهش قهوهای شدن کالوس و تولید ذیتوده آن در دو گونه از شیرینبیان Glycyrrhiza glabra و G. uralensis
DOR: 98.1000/1735-0891.1398.27.131.53.1.1576.115 ریشهها و استولنهای شیرینبیان یکی از مهمترین داروهای خام را در دنیا تشکیل میدهند و محتوای مقادیر زیادی از ماده مؤثر گلیسیریزین، که یک نوع تریترپنوئید ساپونین است، میباشند. کشت بافت و تولید کالوس در شیرینبیان پیشنیاز مطالعات بررسی متابولیتهای ثانویه و ترانسفورماسیون سلولی در این گیاه میباشد. بهمنظور تأثیر آنتیاکسیدانها در کاهش قهوهای شدن و رشد کالوس در دو گونه از شیرینبیان Glycyrrhiza glabra و G. uralensisاز اسید اسکوربیک و PVP (پلیونیل پیرولیدین) در چهار سطح مختلف استفاده شد. همچنین بهمنظور تأثیر این آنتیاکسیدانها در شاخص رشد کالوس، اقدام به کشت سوسپانسیون سلولی در هر دو گونه گردید. برای تولید کالوس از هیپوکوتیل گیاهچههای دو هفتهای و هورمونهای NAA (نفتالین استیک اسید) به غلظت نیم میلیگرم در لیتر و BA (بنزیل آمین) به غلظت دو میلیگرم در لیتر به همراه تیمارهای آنتیاکسیدانهای اسید اسکوربیک و PVP در غلظتهای مختلف در قالب دو آزمایش فاکتوریل جداگانه بر پایه طرح کاملاً تصادفی استفاده شد. نتایج نشان داد در واکشتهای متوالی میزان 1200 میلیگرم در لیتر PVP در هر دو گونه، کمترین باززایی، نکروزه و بیشترین میزان کالوس را دارا بود. درحالیکه در کشت سوسپانسیون سلولی با افزایش غلظت PVP شاخص رشد کالوس کاهش یافته و تیمار PVP با غلظت 1200 میلیگرم در لیتر در هر دو گونه کمترین شاخص رشد کالوس را داشت، که میتواند به دلیل جذب هورمونها (NAA و BA) توسط هورمون PVP و کاهش غلظت هورمون در محیط باشد. تیمار اسید اسکوربیک بهترتیب با غلظتهای 80 و 100 میلیگرم در لیتر در هر دو گونه بیشترین شاخص رشد کالوس را داشت و برای کشت سوسپانسیون سلولی توصیه گردید.

بررسی نوع محیط کشت و تنظیمکننده های رشد گیاهی بر باززایی درونشیشهای گیاه دارویی و مرتعی اروانه یزدی (Hymenocrater yazdianus)
DOR: 98.1000/1735-0891.1398.27.144.53.1.1576.41 اروانه یزدی (Hymenocrater yazdianus) از گونه های مهم دارویی و مرتعی متعلق به تیره نعنا میباشد. در این تحقیق تکثیر انبوه این گیاه با روش کشت بافت بررسی شد. پس از ضدعفونی جوانههای جانبی و انتهایی به عنوان ریزنمونه در کلرید جیوه 1/0 درصد، سه بار شستوشو با آبمقطر استریل حاوی اسید سیتریک به غلظت 1/0 درصد، در محیط کشت موراشیگ و اسکوگ (MS) (1962) بدون تنظیمکننده رشد گیاهی استقرار یافت. سپس گیاهچهها برای انجام پرآوری به همین محیط کشت، شامل ترکیب چهار غلظت بنزیل آدنین (BA) (0/0، 5/0، 1، 5/1 میلیگرم بر لیتر) و سه سطح ایندول بوتیریک اسید (IBA) (0/0، 01/0 و 1/0 میلیگرم بر لیتر) در 12 تیمار و 8 تکرار منتقل شدند. نتایج نشان داد که بیشترین شاخه زایی در محیط کشت موراشیگ و اسگوگ با غلظت 1 میلیگرم بر لیتر بنزیل آدنین و 01/0 میلیگرم بر لیتر ایندول بوتیریک اسید با متوسط 5/5 شاخه به دست آمد. برای ریشه زایی محیطهای کشت موراشیگ و اسگوگ و لینسمایر و اسگوگ (LS) (1956) مورد بررسی قرار گرفتند. در این محیط ها از پنج سطح ایندول بوتیریک اسید (0، 05/0، 1/0، 1 و 2 میلیگرم بر لیتر) استفاده شد و 95 درصد گیاهچه ها در محیط کشت موراشیگ و اسگوگ با 2 میلیگرم بر لیتر ایندول بوتیریک اسید ریشهدار شدند. در نهایت، 83 درصد گیاهچههای ریشه دار شده در محیط پیت و پرلیت با نسبت 2 به 1 با موفقیت سازگار گردیدند.

بهینهسازی باززایی کاکوتی کوهی (Ziziphora clinopodioides Lam)
DOR:98.1000/1735-0891.1398.27.144.53.1.1576.41 گیاه دارویی و معطر کاکوتی کوهی با نام علمی Ziziphora clinopodioides L. متعلق به جنس Ziziphora و تیره نعناع میباشد. هدف از این مطالعه بهینهسازی باززایی این گیاه میباشد و برای این کار از دو کموتایپ با کدهای 5 و 6 استفاده شد. بدینمنظور، ابتدا آزمون برای کنترل آلودگی از هیپوکلریت سدیم یک درصد و الکل 96% استفاده شد. در مرحله بعد کنترل ترکیبات فنولی تولید شده با استفاده از ترکیبات سه ماده PVP (پلیوینیل پیرولیدین)، زغال فعال و اسید آسکوربیک بررسی شد. شاخهزایی چندگانه با استفاده از ریزنمونه جوانه جانبی در محیط کشت MS حاوی ترکیب هورمونی BAP در پنج سطح در ترکیب با NAA (نفتالین استیک اسید) با غلظت 01/0 میلی گرم در لیتر انجام گردید. ریشهزایی شاخه ها در محیط کشت MS حاوی ترکیب هورمونی NAA در دو غلظت 1/0 و 3/0 میلی گرم در لیتر انجام شد. نتایج نشان داد که تیمار هیپوکلریت سدیم یک درصد بههمراه الکل 96 % موفق به رفع آلودگی تا 74 درصد شد. تیمار ترکیب پلیوینیل پیرولیدین، اسید آسکوربیک و زغال فعال با میانگین 25/8 درصد قهوهای شدن در هر دو کموتایپ 6 و 5 کمترین میزان قهوهای شدن را نشان داد. بیشترین شاخه زایی در کموتایپ 5 با 88 درصد در تیمار 2 میلیگرم در لیتر BAP به اضافه 01/0 میلی گرم در لیتر NAA و در کموتایپ 6 با 100 درصد در تیمار 5/2 میلیگرم در لیتر BAP به اضافه 01/0 میلیگرم در لیتر NAA بهدست آمد. بیشترین درصد ریشه زایی در کموتایپ 6، در تیمار 3/0 میلیگرم در لیتر NAA با 3/33 درصد مشاهده شد. نتایج این پژوهش میتواند در ریزازدیادی و تکثیر وسیع این گیاه استفاده شود.

ارزیابی تنوع مورفولوژیکی برخی جمعیتهای زیره سیاه ایرانی (Bunium persicum) در استان خراسان
DOR: 98.1000/1735-0891.1398.27.167.53.1.32.1606 بهمنظور بررسی تنوع فنوتیپی زیره سیاه ایرانی (Bunium persicum) هفت جمعیت از این گیاه در رویشگاه های طبیعی آن در استانهای خراسان رضوی و جنوبی بررسی شدند. نتایج نشان داد تنوع زیادی در بین جمعیتها بهویژه از نظر صفات تعداد چترک های چتر، طول برگ، تعداد بذر در بوته و عرض گل آذین وجود داشت. تجزیه خوشه ای نیز جمعیتها را به 2 گروه تقسیم کرد و نشان داد که تنوع مورفولوژیکی با تنوع جغرافیایی مطابقت دارد. ارتفاع بوته با تعداد چتر در بوته، طول برگ، طول و عرض گل آذین و همچنین طول گل آذین با عرض گل آذین نیز همبستگی مثبت بالایی با یکدیگر داشتند. تجزیه به مؤلفه ها نشان داد که 4 مؤلفه اصلی توانستند 81/93% از کل واریانس صفات را توجیه نمایند که مؤلفه اول با بیشترین سهم (08/52%) شامل صفات ارتفاع بوته، تعداد چتر در بوته، تعداد چترک در چتر اصلی، طول برگ، طول و عرض گل آذین و تعداد بذر در بوته بود. طبق نتایج بدستآمده از این تحقیق مشخص شد که سه جمعیت دهبار، دره ارغوان و فریزی چناران به دلیل داشتن صفات مطلوبی ازجمله ارتفاع بوته بلند، طول برگ بیشتر، تعداد چترک در چتر و تعداد بذر در بوته بالا گیاهان مناسبی برای کارهای اصلاحی و اهلی سازی می باشند.
