باززایی غیرمستقیم کیکم ترکمنی (Acer monospessulanum) در شرایط درون شیشهای
گونه کیکم یکی از گونه های مهم جنس Acer از خانواده Aceraceae میباشد. زیرگونه turcomanicum در جنگلهای ناحیه رویشی ایران تورانی بهعنوان گونـه همـراه ارس در ارتفاعات استانهـای خراسـان رضـوی و شـمالی پراکنش دارد. سال بذردهى این گونه هر سه سال یکبار است. قوه نامیه بذر آن پایین و تعداد بذرهای رسیده و سالم کم میباشد. بذرهای این گونه دارای خواب بوده و قوه نامیه آن پس از طى دوره سرمادهى کاهش مییابد. متأسفانه، بسیاری از رویشگاههای این گونه تحت تأثیر چرای دام و دخالتهای انسانی با تخریب زیادی مواجه شده است. بهدلیل مشکلات تکثیر جنسی، استفاده از روشهای غیرجنسی بهمنظور تولید نهالهای با کیفیت مطلوب میتواند در احیاء رویشگاههای این گونه بسیار راهگشا باشد. یکی از راههای مطمئن تولید نهال، تکثیر غیرجنسی از طریق ریزازدیادی است. به این منظور ریزنمونهها از رویشگاه آن تهیه و باززایی انجام شد. القاء کالوس در محیطهای هورمونی مختلف انجام و غلظتهای مختلف هورمونی برای باززایی کالوس بررسی شد. از بین محیطهای مورد بررسی، باززایی غیرمستقیم از کالوس تنها در غلظتهای 1/0 و 5/0 میلیگرم در لیتر هورمون رشد گیاهی تیدیازورون (TDZ) انجام شد. تعداد، رشد طولی و شادابی ساقهچهها تحت تأثیر غلظتهای مختلف هورمون TDZ بررسی شد و تیمار 5/0 میلیگرم در لیتر هورمون TDZ با متوسط 16/4 عدد بیشترین تعداد ساقهزایی را داشت. از نظر شادابی و ارتفاع ساقهچهها تفاوت معنیداری بین تیمارها وجود نداشت. بهطورکلی محیط پایه MS و هورمون TDZ برای باززایی کالوس گونه کیکم ترکمنی مناسب میباشد. گیاهچههای باززایی شده بهمنظور سازگاری به گلدان منتقل و بعد به خاک انتقال یافتند.

تأثیر نور و تنظیمکننده رشد در تکثیر بادامکوهی (Amygdalus scoparia) به روش کشت درونشیشهای جنین
بادام کوهی (Amygdalus scoparia Spach) یکی از گونههای ارزشمند بادام با کابردهای زیستمحیطی، حفظ خاک، دارویی و خوراکی است که بهعنوان پایه در توسعه کشت بادام اهلی نیز مورد استفاده قرار میگیرد. در این پژوهش پس از استقرار درونشیشهای جنین بادام کوهی و رشد شاخسارهها در محیط کشت DKW با ترکیب تنظیمکنندههای رشد IBA، BA و TDZبا مقادیر بهترتیب 01/0، 5/0 و 05/0 میلیگرم در لیتر، تأثیر عوامل نور و تنظیمکنندههای رشد بر ریشهزایی شاخسارههای حاصل از جنین در یک آزمایش فاکتوریل در قالب طرح پایه کاملا تصادفی مورد مطالعه قرار گرفت. از نور بهعنوان عامل اول در دو سطح (الف: 16 ساعت روشنایی و 8 ساعت تاریکی، ب: دائم در تاریکی به مدت 10 روز) و تنظیمکننده رشد در محیط کشت پایه DKW بهعنوان عامل دوم در پنج سطح (1: NAA + IBA هریک به میزان 1 میلیگرم در لیتر، 2: NAA به میزان 1 میلیگرم در لیتر، 3: IBA به میزان 1 میلیگرم در لیتر، 4: IBA به میزان نیم میلیگرم در لیتر، 5: NAA به میزان نیم میلیگرم در لیتر) استفاده شد. براساس نتایج تجزیه واریانس، دو عامل نور و تنظیمکننده رشد تأثیر معنیدار و متفاوتی بر همه صفات مورد مطالعه در ریشهزایی داشتند. روشنایی موجب رشد بهتر شاخسارهها نسبت به تاریکی شد. سطوح مختلف تنظیمکنندههای رشد بهکار رفته در محیط کشت هم اثرات متنوعی بر رشد شاخسارهها و ریشهزایی گذاشتند. ترکیب IBA و NAA بیشترین تعداد ریشه را تولید کرد. ولی IBA به مقدار 1 میلیگرم در لیتر بیشترین طول ریشه اصلی، تعداد و طول ریشههای فرعی را ایجاد کرد. اثر متقابل عوامل نور و محیط کشت بر طول شاخساره، طول ریشه اصلی، تعداد و طول ریشه فرعی اثر معنیداری گذاشت، بهطوریکه ترکیب دو تنظیمکننده رشد در روشنایی بیشترین تعداد ولی استفاده از 1 میلیگرم NAA در تاریکی بیشترین تعداد ریشه فرعی را تولید کرد. بر اساس نتایج حاصل از این مطالعات، وجود تناوب نور و تاریکی و استفاده از تنظیمکننده رشد IBA در تکثیر رویشی ژنوتیپهای مختلف بادام کوهی ضروریست.

اثرات القاء پلیپلوئیدی در شرایط درون شیشهای بر برخی خصوصیات مورفولوژیکی، فیزیولوژیکی و بیوشیمیایی گیاه نوروزک ((Salvia leriifolia Benth.
نوروزک (.Salvia leriifolia Benth) گیاهی علفی و چندساله، متعلق به تیره نعناعیان که در سالهای اخیر خواص مختلف دارویی این گیاه شناخته شده است. پلیپلوئیدی درون شیشهای گیاهان دارویی در سالهای اخیر یکی از موضوعات جالب توجه در بیوتکنولوژی و کشت بافت گیاهی میباشند. بر این اساس، با استفاده از غلظتهای مختلف کلشیسین (0، 05/0، 1/0، 2/0 و 5/0 درصد) در سه سطح زمانی 24، 48 و 72 ساعت، القای پلیپلوئیدی در جوانههای انتهایی گیاهچههای باززا شده نوروزکمطالعه شد. برای شناسایی گیاهان پلیپلوئید، خصوصیات مورفولوژیکی، فیزیولوژیکی، بیوشیمایی و سیتوژنتیکی بررسی شد. نتایج نشان داد کهتأثیر تیمارهای مختلف کلشیسین بر درصد زندهمانی گیاهچههای حاصل در سطح احتمال 1 درصد معنیدار میباشد. با مقایسه تیمارهای 05/0، 1/0، 2/0 و 5/0 درصد کلشیسین، بالاترین میزان زندهمانی ریزنمونهها در تیمار 05/0 درصد برابر با 44/84 درصد مشاهده شد. همچنین بالاترین میزان مرگ گیاهچه، در غلظت 5/0 درصد کلشیسین ثبت شد. القاء تتراپلوئیدی سبب تغییرات مورفولوژیکی، فیزیولوژیکی، سیتولوژیکی و ترکیبات شیمیایی از قبیل افزایش طول و عرض روزنه در سطح زیرین برگ، عرض برگ، سطح برگ، میزان کلروفیل برگ، میزان آنتیاکسیدان کل، فنل کل و فعالیت آنزیمهای آنتیاکسیدانی شد (آنزیمهای کاتالاز و آسکوربات پراکسیداز)، همچنین موجب کاهش طول برگ و تراکم روزنه در واحد سطح برگ شد. تعداد کروموزوم گیاهان دیپلوئید برابر با 22 عدد (2n=2x=22) و در گیاهان تتراپلوئید برابر با 44 (2n=4x=44) بود. بنابراین میتوان بیان کرد که کلشیسین بهطور مؤثری قابلیت القای پلیپلوئید در گیاه نوروزک را دارد.

واکنش متفاوت دورگهای بین گونهای صنوبر به تکثیر از طریق ریزازدیادی
گونههای مختلف صنوبر قابلیتهای متفاوتی دارند که آنها را از هم متمایز میکند. ولی این تفاوتها موجب شده است که برخی از گونهها در شرایط متضاد اقلیمی سازگاری پیدا کنند. دورگهای بین این گونهها نیز قابلیتهای بینابینی از خود نشان میدهند که ممکن است بتوانند دامنه اکولوژیک صنوبرها را گسترش دهند. در این تحقیق سه پایه از دورگهای بین گونهای حاصل از تلاقی بین دو گونه از صنوبر بهنامهای سپیدار (Populus alba)و پده(Populus euphratica) با ریزازدیادی تکثیر شدند تا ضمن بهینهکردن روش تکثیر این دورگها، تفاوتهای احتمالی واکنش بهریزازدیادی در آنها نیز بررسی شود. در بخش پرآوری دورگهای مورد مطالعه واکنشهای متفاوتی بهمحیط کشتهای مورد استفاده از خود نشان دادند، بهطوریکه یکی از دورگها در سطوح سهگانه محیطهای پرآوری، بهمیزان زیادی ریشه هم تولید کرد. این دورگ در مرحله پرآوری از نظر صفاتی مانند تعداد شاخه، طول شاخه، تعداد جوانههای جانبی و مقدار ریشه برتری زیادی نسبت به سایر پایهها از خود نشان داد. در مرحله ریشهزایی هم پایه برتر در مرحله تکثیر، برتری ویژهای از نظر تعداد و طول ریشه از خود نشان داد. سهولت در ریشهدهی یکی از دورگها، که در صورت انتخاب بهعنوان یک رقم تجاری باید بهصورت رویشی تکثیر و توسعه داده شود، میتواند امتیاز ویژهای برای این دورگ باشد. البته اثر متقابل موجود بین دو عامل مورد مطالعه هم در بیشتر موارد برتری یکی از دورگها را نشان داد.

تأثیر شدت خشکیدگی تاجی بر برخی ویژگیهای فیزیولوژیک درختان بلوط ایرانی (Quercus brantii Var Persica)
در این پژوهش اثر شدت خشکیدگی تاجی درختان بلوط ایرانی بر خصوصیات فیزیولوژیک برگ و شاخه در جنگل دچار خشکیدگی ملهسیاه استان ایلام در فصول بهار و تابستان سالهای 1393-1392 پایش شد. درختان بلوط بر اساس شدت خشکیدگی تاجی به چهار گروه با تعداد شش تکرار تقسیم شدند. نمونهگیری از برگ و شاخه دوساله درختان بلوط در جهت جنوبی تاج آنها بهطور تصادفی انجام و نمونهها برای اندازهگیری آنزیمهای پراکسیداز و کاتالاز، پرولین، کلروفیل، کارتنوئید و محتوای رطوبت نسبیبرگ به آزمایشگاه منتقل شدند. نتایج بررسی اثر شدت خشکیدگی تاجی بر صفات مورد مطالعه نشان داد که محتوای رطوبت نسبیبرگ درختان سرخشکیده بیشتر از درختان سالم است. میزان پرولین برگ درختان سرخشکیده بیشتر از درختان سالم بود. سایر صفات مورد مطالعه تغییر معنیداری بین درختان سالم و سرخشکیده نشان ندادند. نتایج بررسی اثر متقابل سال و فصل بر صفات مورد مطالعه نشان داد که پراکسیداز شاخه در بهار 93 بیشترین میزان را داشت، در بهار و تابستان 92 کمتر بود و در تابستان 93 کمترین مقدار را داشت. کاتالاز برگ در بهار و تابستان 92 بیشترین میزان و در بهار 93 کمترین میزان بود. کاتالاز شاخه در بهار 92 بیشترین میزان و در بهار و تابستان 93 کمترین میزان را داشت. نتایج نشان داد که تغییرات سالیانه (از 1392 تا 1393) میزان کلروفیل b، کلروفیل کل و محتوای رطوبت نسبی برگ بهصورت افزایشی و کارتنوئید بهصورت کاهشی بود. نتایج نشان داد که تغییرات فصلی (از بهار تا تابستان) میزان کلروفیل a، کارتنوئید و محتوای رطوبت نسبی برگ بهصورت افزایشی، پرولین در سال اول تحقیق بهصورت کاهشی و در سال دوم تحقیق بهصورت افزایشی بود. نتیجهگیری شد که تغییرات مقادیر محتوای رطوبت نسبی برگ و پرولین در درختان سرخشکیده بلوط ایرانی در جهت کاهش تنش در پیکره درخت و گذر از بحران خشکسالی و عدم تغییرات معنیدار رنگیزههای فتوسنتزی و آنزیمها در جهت حفظ و تداوم فعالیتهای حیاتی در پیکره آن است.

ارزیابی تنوع ژنتیکی عملکرد دو گونه مرزه Satureja mutica و Satureja isophylla در شرایط دیم دماوند
عملکرد گونههای Satureja muticaو S. isophyllaو پنجاکسشن از گونهS. muticaدر شرایط دیم منطقه دماوند در قالب طرح بلوکهای کامل تصادفی با سه تکرار ارزیابی شد. گونههااختلاف معنیداری برای وزن خشک بوته، درصد اسانس، قطر تاج پوشش گیاه، ارتفاع بوته، تعداد شاخههای فرعی و طول برگ داشتند و گونه S.muticaاز نظر این صفات برتر از گونه S. isophylla بود. همچنین، اکسشنهای گونه S.muticaبرای بازده اسانس، طول و عرض برگ، ارتفاع گیاهو تعداد شاخه اصلی و فرعی تفاوت معنیدار نشان دادند ودو اکسشن از شمالشرقی کشور بهترتیب برای بازده اسانس و تعداد شاخه و یک اکسشن از شمال کشور از نظر ارتفاع گیاه و طول برگ دارای بالاترین میزان بودند. همچنین، وزن خشک بوته با قطر تاجپوشش و تعداد شاخه همبستگی مثبت و معنیداری نشان دادند. در تجزیه به مؤلفههای اصلی، 3 مؤلفه اول بهترتیب با 66، 20 و 9 درصد و در مجموع بیش از 95 درصد از تنوع متغیرها را تبیین کردند. در مؤلفه اول، قطرتاجپوشش، وزن خشک بوته و میزان اسانس، در مؤلفه دوم، ابعاد برگ و در مؤلفه سوم، تعداد شاخههای اصلی و فرعی مؤثرترین صفات در این تغییرات بودند. تجزیه خوشهای، گونه S. isophyllaرادریک گروه مجزا قرار داد. اکسشنهای mu1Kh و mu2Kh گونه S. muticaدر یک گروه و اکسشنهای mu2Gl ، mu3Gl و mu1Mz این گونه در گروه دیگر قرار گرفتند. باتوجه به نتایج این مطالعه، اکسشن mu1Khمرزه S.muticaقابل کشت و بهرهبرداری در شرایط دیم منطقه دماوند میباشد.

ارزیابی روابط کاریوتیپی برخی گونههای بخش های Bromus و Genea از جنس بروموس
جنس بروموس از مهمترین گیاهان مرتعی میباشد که در سطح وسیعی از عرصههای طبیعی کشور پراکنش دارد. روابط بین گونهای 45 جمعیت دیپلوئید از 9 گونه متعلق به دو بخش Genea و Bromus بههمراه 5 جمعیت از گونه چچم یکساله بر اساس خصوصیات کاریوتیپی بررسی شد. در هر جمعیت 5 سلول متافازی از مریستم انتهایی ریشه بذرهای جوانهزده تهیه شد. طول کل کروموزوم، طول بازوی بلند و کوتاه اندازهگیری شد. شاخص سانترومری و نسبت بازوها، محتوای نسبی کروماتین، شاخص نامتقارن بودن درون و بین کروموزومی و درصد شکل کلی کروموزوم محاسبه شد. تقارن کاریوتیپی تعیین و نوع کروموزومها مشخص شد. تجزیه واریانس اختلاف معنیداری را در سطح یکدرصد در خصوصیات کاریوتیپی در بین گونهها و جمعیتهای مورد مطالعه نشان داد. جمعیتها کاریوتیپ متقارنی داشتند و بر اساس جدول دوطرفه استبینز در کلاس A1 و A2 قرار گرفتند و دارای کروموزومهای متاسنتریک و تعداد کمی کروموزوم ساب متاسنتریک بودند. گونههای دو بخش براساس تجزیه به مؤلفههای اصلی و تجزیه خوشهای تفکیک شدند. تکامل کاریوتیپی گونههای بخش Bromus بیشتر از طریق مقدار کروماتین و بخش Genea بیشتر از طریق افزایش عدم تقارن درون و بین کروموزومی بود. در بخش Bromus دو گونهBromus briziformis وB. squarrosus بیشترین و دو گونهB. scoparius و B. danthoniae کمترین تکامل کاریوتیپی را داشتند. از طرف دیگر دو گونهB. rechingeri و B. japonicus در بین این دو گروه قرار گرفتند. برای بخش Genea نیز گونه B. tectorum تکامل کاریوتیپی بیشتری نسبت به دو گونه B. sterilis و B. sericeus نشان داد.

گل انگشتانه از منابع مهم ترکیبات دارویی گلیکوزیدهای قلبی مانند دیگوکسین، دیژیتوکسین و لاناتوزیدها میباشند که در نارسایهایی احتقانی قلب کاربرد درمانی دارند. جنس Digitalis دارای چندین گونه با ارزش است و تنها گونه بومی ایران گونه نروسا (Digitalis nervosa) میباشد که تعدادی گلیکوزید قلبی در آن شناسایی شده است. نظر به اهمیت مطالعه و بررسی گیاهان بومی بهمنظور استفاده در صنایع دارویی، این پژوهش بر روی گونه نروسا انجام شد. یکی از ژنهای کلیدی مهم در مسیر بیوسنتز کاردنولیدها در گیاه گل انگشتانه ژن 3β-HSDمیباشد که کدکننده آنزیم 3β-HSD است که در مراحل اولیه بیوسنتز کاردنولیدها نقش کلیدی دارد. هدف این تحقیق توالییابی، بررسیهای فیلوژنتیکی و بیان ژن 3β-HSD در بافتهای مختلف و تحت تیمار با الیسیتورهای سالیسیلیک اسید و متیل جاسمونات بود. نتایج بهدست آمده آشکار کرد که توالی کدکننده این آنزیم با گونه Digitalis lanata قرابت بسیار نزدیکی دارد. در بررسی بیان ژن 3β-HSD در سطح رونوشت نیز مشخص شد که در بافت برگ دارای میزان بیان چشمگیری است و تحت تیمارهای متیل جاسمونات و سالیسیلیک اسید میزان بیان این ژن افزایش مییابد. نتایج حاصل از این پژوهش میتواند در راستای افزایش کاردنولیدها از طریق مهندسی متابولیک و دستورزی ژنتیکی مفید واقع شود.

بررسی ساختار تنوع ژنتیکی جمعیتهای نیای وحشی گندم (Aegilops crassa) با استفاده از نشانگرهای بین ریزماهواره ژنومی
بهمنظور بررسی تنوع ژنتیکی 16 جمعیت از گونه Aegilops crassa با استفاده از 10 آغازگر ISSR، در مجموع 105 آلل تکثیر شدند که از این تعداد، 86 آلل (90/81 درصد) بهعنوان آلل چندشکل تشخیص داده شدند. تعداد آللهای تکثیر شده از 6 تا 15 با میانگین 11 آلل متغیر بود. محتوای اطلاعات چندشکلی از 17/0 در آغازگر UBC842 تا 34/0 برای آغازگر ISSR12 متفاوت بود. همچنین شاخص نشانگر از 98/0 برای آغازگر UBC842 تا 7/2 برای آغازگر ISSR08 متفاوت بود. میانگین شباهت ژنتیکی محاسبه شده برای اطلاعات نشانگرهای بین ریزماهوارهای برابر 89/0 بود و از 76/0 (بین دو ژنوتیپ از کرمانشاه و تبریز) تا 96/0 (بین دو ژنوتیپ از ایلام و کرمانشاه) متفاوت بود. تجزیه خوشهای بر اساس دادههای مولکولی، ژنوتیپها را در سه گروه جداگانه قرار داد که بهوسیله تجزیه واریانس مولکولی تأیید شد. البته روشهای گروهبندی خوشهای و تجزیه به مختصات اصلی نتوانست جمعیتها را بهطور کامل از هم تفکیک کند و عدم ارتباط بین تنوع مولکولی و تنوع جغرافیایی را نشان داد که نشاندهنده تنوع ژنتیکی بالای این جمعیتها میباشد. تجزیه واریانس مولکولی نشان داد که سطح بیشتری از تنوع به درون جمعیتها (53 درصد) تعلق داشت، درحالیکه (47 درصد) تنوع در بین جمعیتها مشاهده شد.

اثر مبدأ بذر و پایه مادری بر جوانهزنی بذر و رشد نونهالهای کیکم (مطالعه موردی: جنگل چنارناز در جنوب استان یزد)
بهمنظور بررسی تأثیر مبدأ بذر و پایه مادری بر روی میزان جوانهزنی و برخی خصوصیات مورفولوژیکی نونهالهای کیکم (Acer monspessulanum)، بذر آن از 20 پایه مادری در دو جمعیت (با ارتفاع از سطح دریای 2400 و 2700 متر) واقع در جنگل چنارناز در جنوب استان یزد جمعآوری شد و بعد در گلخانه تحقیقاتی واقع در دانشگاه یزد در قالب طرح کاملاً تصادفی در سه تکرار کاشته شد. نتایج تجزیهوتحلیل واریانس صفات مورد مطالعه نشان داد که تأثیر ارتفاع از سطح دریای مبدأ بذر بر روی صفات قطر یقه، طول ریشه و وزن تر ریشه نونهالها معنیدار بود، بهطوریکه در بین این مشخصهها، قطر یقه و طول ریشه نونهالهای حاصل از رویشگاه با ارتفاع 2700 متر از سطح دریا، از وضعیت مطلوبتری نسبت به سایر نهالها برخوردار بودند. هر چند تا حدودی رویشگاه با ارتفاع 2700 متر با توجه به خصوصیات مورد بررسی نونهالهای پایههای مادری آنها بهمنظور جمعآوری بذر و تولید نهال و جنگلکاری و متعاقب آن احیای عرصههای تخریب یافته، مناسب تشخیص داده شد اما درصد کمتری از پایههای مادری رویشگاه با ارتفاع 2400 متر نیز میتوانند بهمنظور حفظ تنوع مورد توجه قرار گیرند. از اینرو در هر جمعیت، پایههای برتر از لحاظ مقدار عددی صفات، مشخص شد که پیشنهاد میشود توجه ویژهای به این پایهها به منظور جمعآوری بذر، معطوف شود.

بررسی تنوع ژنتیکی اکوتیپهای مریم نخودی (Teucrium polium) با استفاده از نشانگر مولکولی ISSR
مریم نخودی گیاهی است از خانواده نعناعیان و کاربرد وسیعی در صنعت داروسازی دارد. تنوع ژنتیکی 17 اکوتیپ مریم نخودی جمعآوری شده از سراسر استان کرمانشاه با استفاده از نشانگر مولکولی ISSR مورد بررسی قرار گرفت. از 15 آغازگر ISSR استفاده شد که از میان آنها 12 عدد دارای نوارهای قابل امتیازدهی بودند. آغازگرهای ISSR در مجموع توانستند 82 نوار تولید کنند، که 80 باند چند شکل مشاهده شد. میانگین تعداد نوار تولید شده توسط هر آغازگر برای 17 اکوتیپ برابر 83/6 بود. آغازگر IS6 بیشترین تعداد نوار (12 نوار) و آغازگر IS3 کمترین تعداد نوار (4 نوار) را نشان دادند. نشانگر ISSR در بین اکوتیپها چندشکلی مطلوبی نشان داد و تمام آغازگرها برای بررسیهای گونه مریم نخودی مناسب بودند. نتایج نشان داد که اکوتیپ 1G (صحنه) بیشترین فاصله ژنتیکی را با اکوتیپهای 12 G(صحنه)، 7 G(گیلان غرب)، 13G (ثلاث) و 8G (دالاهو) داشت. گروهبندی حاصل از تجزیه خوشهای نشان داد که اکوتیپها در چهار گروه قرار گرفته و تنوع ژنتیکی با تنوع جغرافیایی تطابق نداشت. همچنین نتایج حاصل از گروهبندی با استفاده از تجزیه واریانس مولکولی (AMOVA) و تجزیه به مختصات اصلی (PCo) تأیید شد.

مطالعه اثرات ترکیب محیطکشت و نوع ریزنمونه در بهینهسازی کشت خارشتر (Alhagi camelorum F.)
خارشتر (F. Alhagi camelorum) گیاهی است از خانواده بقولات که سازگاری خوبی با شرایط آب و هوایی مناطق خشک دارد. به دلیل پروتئین قابل هضم بالا، همچنین امکان بهرهبرداری ترنجبین از ساقههای آن و سازگاری با شرایط سخت زیستمحیطی به عنوان یکی از گونههای مهم مرتعی و دارویی محسوب میشود. تکثیر این گونه در طبیعت توسط بذر و ریزوم انجام میشود، ولی بذر آن برای جوانهزنی دارای مشکل است. هدف این تحقیق، بررسی تأثیر غلظتهای مختلف ساکارز در ریزازدیادی گیاه خارشتر از طریق کشت تخمدان و جنین بذری برای بهبود تکثیر آن میباشد. برای این هدف، با جداسازی جنین از تخمدان و همچنین حذف پوسته بذر (جنینبذری)، ریزنمونهها در محیطکشت MS حاوی غلظتهای 0، 15، 30، 45 و 60 گرم در لیتر ساکارز برای بررسی جوانهزنی و تکثیر کشت شدند. درصد جوانهزنی و سایر صفات رشد گیاهچه طی چهار هفته بررسی شد. نتایج نشان داد که بین دو ریزنمونه در میزان درصد جوانهزنی و طول گیاهچه اختلاف معنیدار وجود داشت و درصد جوانهزنی جنینبذری در مقایسه با تخمدان بهطور معنیداری (P<0.01) بیشتر بود. بیشترین درصد جوانهزنی جنین بذری (3/93 درصد) در تیمار 15 گرم در لیتر ساکارز بهدست آمد که با مقایسه با سایر غلظتهای ساکارز اختلاف معنیدار و مثبت داشت. با افزایش غلظت ساکارز تا 15 گرم بر لیتر سایر صفات رشد جنین مانند طول ریشهچه، طول ساقهچه و همچنین شاخص بنیه افزایش یافت. البته غلظتهای بیشتر از 15 گرم در لیتر ساکارز در تمامی صفات اندازهگیری شده جنین اثر منفی داشت.

آویشن باغی (Thymus vulgaris) یکی از مهمترین گیاهان دارویی است که بیش از 20 نوع ترکیب شیمیایی مهم شناخته شده دارد. ژن 1-دی اکسی دی زایلوز 5-فسفات ردوکتوایزومراز (DXR) بهعنوان نقطه کنترلی مهم در مسیر MEP عمل میکند، زیرا اولین مرحله اصلی و متمایزکننده مسیر MEP میباشد. گاماترپینن جزء کلیدی اسانس در آویشن باغی میباشد، که حدود 30% از اسانس را تشکیل میدهد و بهوسیله گاماترپینن سنتاز تولید میشود. سیتوکرومهای مختلف از P450s شامل CYP71D178 و CYP71D180 است که با واسطه گاماترپینن در بیوسنتز تیمول و کارواکرول دخیل هستند. این پژوهش، با هدف بررسی الگوی بیان ژنهای دخیل در بیوسنتز تیمول و کارواکرول شامل DXR، گاماترپینن سنتاز، CYP71D178وCYP71D180در بافتهای گل و برگ آویشن باغی انجام شد. در این مطالعه الگوی بیان این ژنهادر بافتهای گل و برگ با استفاده از تکنیک RT-PCR نیمهکمیبررسی شد. میزان بیان ژن در بافت گل نسبت به برگ برای ژنهای DXR، گاماترپینن سنتاز، CYP71D178 و CYP71D180 بهترتیب 7/1، 7/2، 9/1 و 2 برابر بیشتر بود و شدت بیان ژنهای مختلف نیز با هم متفاوت بود. میزان بیان این ژنها در کرکهای غدهای بالا بود. بنابراین بیان بالا در بافت گل احتمالاً بهدلیل تراکم بالای کرکهای غدهای میباشد. با توجه به این نتایج میزان اسانس بیشتر در گل نسبت به برگ میتواند ناشی از بیان بیشتر ژنهای بررسی شده در گل نسبت به برگ باشد.

ارزیابی تنوع مورفولوژیکی برگ و میوه جمعیتهای طبیعی گلابی وحشی (Pyrus glabra Boiss.) در جنگلهای زاگرس جنوبی
بهمنظور بررسی تنوع مورفولوژیکی برگ و میوه جمعیتهای مختلف گلابی وحشی (Pyrus glabra)، نمونههای برگ و میوه تعداد 48 پایه از چهار جمعیت از این گونه در ارتفاع 2150 متر از سطح دریا (تنگسرخ)، 2250 متر (وزگ)،2450 متر (گرگو) و 2550 متر (ماهپرویز) واقع در حوزه شهرستان بویراحمد نمونهبرداری شد. سپس 16 صفت کمی و کیفی مربوط به خصوصیات مورفولوژیک برگ و میوه اندازهگیری شد. از بین پارامترهای مورد مطالعه، صفات طول پهنک، طول دمبرگ، عرض پهنک در 1/0 و 9/0 طول برگ، حداکثر عرض پهنک، طول نسبی دمبرگ، شکل برگ، سطح برگ و قطر بزرگ و کوچک میوه در بین جمعیتهای مختلف تفاوت معنیداری را با یکدیگر نشان دادند. تجزیه به مؤلفههای اصلی نشان داد که صفاتی مانند حداکثر عرض پهنک، طول پهنک برگ، وزن خشک برگ، زاویه نوک برگ و سطح ویژه برگ بیشترین نقش را در گروهبندی ایفا کردند. همچنین صفات زاویه نوک برگ، وزن خشک برگ و سطح برگ دارای بیشترین ضریب تغییرات بودند. نتایج تجزیهوتحلیل تشخیصی نشان داد که صفات شکل برگ و قطر بزرگ میوه صفات تأثیرگذار در جداسازی جمعیتها بودند. بهطور کلی از صفات شکل قاعده برگ و قطر بزرگ میوه که در برابر تغییرات محیطی تغییرپذیری کمتری دارند میتوان بهعنوان صفات مناسبی برای شناسایی و تفکیک ژنوتیپهای گونه گلابی وحشی استفاده کرد.
